摘    要:目的:构建以绿色荧光蛋白(GFP)为报告基因的酿酒酵母表达载体。方法:利用通用载体质粒融合系统(UPS),首先将GFP片断连入donor载体,随后在Cre酶作用下与acceptor载体融合,获得了带有GFP片断的酿酒酵母(saccharomyces cerevisiae)表达载体。结果:成功构建了以GFP为报告基因的酿酒酵母载体,并在酵母中得到表达。结论:GFP是一种理想的报告基因。同时,利用UPS能够高效、简便地进行酵母表达载体的构建。

Abstract:Objective To construct saccharomyces cerevisiae expression vector with GFP as report gene. Methods By using UPS( univector plasmid - fusion system), GFP gene was linked into donor vector. Saccharomyces cerevisiae expression vector was formed after fusion of donor vector and acceptor vector under the catalyzation of Cre enzyme. Results Saccharomyces cerevisiae expression vector with GFP as report gene was constructed and expressed successfully. Conclusion GFP is an ideal report gene, UPS is efficient and converient for construction of expressing vector.
