失血性休克肠上皮细胞线粒体DNA ATPase 6,8亚基基因的改变

摘    要:目的 观察失血性休克大鼠肠上皮细胞mtDNA ATPase6,8亚基基因的改变。方法 将6只Wistar大鼠分成单纯手术组、休克组。分别提取mtDNA,采用PCR技术扩增后测序,检测突变点。结果 Wistar大鼠mtDNA ATPase6,8亚基基因存在多态性,休克组存在A 7797缺失和一定数量点突变:A7863G,G7950T,C8294G,T8505G。结论 失血性休克5h,肠上皮细胞mtDNA ATPase 6,8亚基基因会发生突变,由此可导致所编码的氨基酸的改变,有可能是酶活性改变的原因。

Abstract:Objective To observe the changes of mitochondrial DNA ATPase 6, 8 subunit genes of small intestine epithelial cells in hemorragic shock rats. Methods Six rats were randomly divided into the control and shock groups, from which mtDNA was extracted, respectively. The sequences were examined after amplification by polymerase chain reaction (PCR) to detect mutation sites. Results The polymorphism existed at some sites of ATPase 6, 8 genes of Wistar rats. There was an A 7797 deletion and several site mutations of ATPase 6,8 subunit genes in shock 5h group, namely, A 7863 G, G 7950 T, C 8294 G and T 8505 G. Conclusion The mutation of ATPase 6, 8 subunit genes at the 5th h may result in the changes of their coding amino acids, which might be due to the changes of F 0F 1 ATPsynthase activity.
