马桑内酯致慢性癫痫鼠海马谷氨酸脱羧酶阳性神经元的变化

摘    要:目的:观察马桑内酯慢性癫痫模型中谷氨酸脱羧酶(glutamlc acid decarhoxylase,GAD)阳性神经元的变化与癫痫的关系。方法:用马桑内酯(coriaria lactone,CL)亚致惊剂量肌注的方法建立慢性癫痫模型,对照组以生理盐水注射。用振动切片机连续冠状切片,选取含海马结构中部的相邻切片,行GAD65/GAD67免疫组织化学染色。结果:1.马桑内酯致痫组大鼠均被点燃,且随着注射次数的增加,发作的潜伏期逐渐缩短(从50min减少到10min左右),发作时间逐渐延长(从数分钟到数十分钟),对照组均无发作。2,海马各区和齿状回GAD65/GAD67阳性神经元数在马桑内酯致痫组明显低于正常组,免疫反应减弱。结论:马桑内酯致痫鼠中GAD65/GAD67神经元数目的减少,免疫反应减弱,与癫痫关系密切。

Abstract:Objective:To observe the relationship between changes of GAD-postive neurons and epilepsy in epileptic model induced by CL.Methods:Chronic epileptic rat models were established by repeated intramuscular injection of CL at subthreshold dosages every 3.5 days, and control group received saline injection instead of CL. During injection seizure latency of rats must be recorded. In each group six rats were used for immunohistochemistry staining of GAD65 and GAD67 respectively. Results: 1) With the increase of the injection times, seizure latency was shortened from 50 mins to 10 mins gradually.2) Compared with control group, numbers of GAD65/GAD67-positive neurons in each region of hippocampus and dentate gyre in epileptic rats decreased significantly. Immunoreactivity also became weakened. Conclusion: It is found that the number of GAD65/GAD67-neruons of hippocampus and dentate gyre decreased in epileptic model induced by CL in comparison with normal control group, and the immunoreactivity was also weakened,and these changes were related to epilepsy.
