复方红景天片对高+Gz应激致大鼠心肌损伤的保护作用研究

摘    要:目的:观察复方红景天片对高正加速度(+Gz)应激致大鼠心肌损伤的保护作用.方法:将84只SD大鼠随机分为空白对照组、应激对照组、高+Gz应激组、阳性药(复方党参片)对照组、复方低剂量组、复方中剂量组和复方高剂量组7组,各组均灌胃给药14 d,第15天进行离心机暴露,观察各组动物心肌酶和心肌超微结构的变化.结果:与空白对照组和应激对照组相比,高+Gz应激组大鼠血清谷草转氨酶(AST)、乳酸脱氢酶(LDH)、肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)明显升高.复方红景天片可以降低高+Gz应激大鼠心肌酶的水平,复方高剂量组4种心肌酶水平与高+Gz应激组相比均明显降低(P<0.05).高+Gz应激组大鼠心肌间质和血管内皮细胞水肿,肌原纤维离散,线粒体变性,闰盘结构解离.复方红景天片组大鼠心肌超微结构后损伤减轻,肌丝结构排列规则,毛细血管结构未见明显改变,复方高剂量组心肌超微结构与对照组接近.结论:复方红景天片对高+Gz应激导致的大鼠心肌损伤有明显的保护作用.

Protective effect of rhodiola crenulata compound on injury of myocardium in rats after high +Gz stress

Abstract:Objective:To observe the protective effect of rhodiola crenulata compound on injury of myocardium in rats after high +Gz stress.Methods:Eighty-four healthy SD rats were randomly divided into seven groups:blank control group,stress control group,high +Gz stress group,positive drug group,low dose of rhodiola crenulata compound group,medium dose of rhodiola crenulata compound group and high dose of rhodiola crenulata compound group,with ten rats in each group.Each rat had been fed 20 mL·kg-1 menstruum once per day for 14 days and was exposed to high +Gz at the 15th day.The level of plasma myocardium enzyme and the changes of myocardial ultrastructure were examined.Results:Compared with those of blank and stress control groups,the plasma levels of myocardium enzyme including AST,LDH,CK and CK-MB in high +Gz stress group were significantly higher.Rhodiola crenulata compound preconditioning could decrease the level of myocardium enzyme and the four myocardium enzymes levels(AST,LDH,CK and CK-MB) in high dose compound group were significantly lower than those in high +Gz stress group(P0.01).The ultrastructural changes of the cardiac muscle were observed in rats of high +Gz stress group,including myocardial interstitial and endothelium edema,myofibril disorder,mitochondrion degeneration and intercalated disc dissociation.The injury of myocardial ultrastructure induced by high +Gz stress relieved in rhodiola crenulata compound preconditioning groups.Myofibril structure was regular and capillary permeability did not show an evident change.The ultrastructure in rats of high dose compound group was essentially normal.Conclusion:Rhodiola crenulata compound had good protective effect on injury of myocardium in rats after high +Gz stress.
