双氧水冲洗对正常脑组织影响的初步探讨

摘    要:目的探讨双氧水冲洗对正常脑组织的影响,对临床使用双氧水冲洗的安全性做出评价。方法以3%双氧水冲洗大鼠脑组织,观察双氧水冲洗后组织细胞和血管的损伤,检测脑组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、糖皮质激素受体表达和血浆中糖皮质激素浓度。结果经3%双氧水冲洗后,大鼠脑组织中MDA含量升高(P〈0.05),SOD活性下降(P〈0.05),脑组织神经细胞、胶质细胞和血管明显损伤,血浆糖皮质激素浓度明显增加(P〈0.05),脑组织糖皮质激素受体表达下降(P〈0.05)。结论经3%双氧水冲洗后,脑组织内有剧烈的脂质过氧化反应和氧化应激反应,说明双氧水对正常脑组织和间质具有较强的损伤作用。在这种损伤过程中血管损伤可能起关键性作用,提示开颅手术应慎用双氧水冲洗脑组织。

Abstract:Objective To study the effect of hydrogen peroxide on brain tissues in order to appraise the safety of imntraoperatively douching brain tissues by hydrogen peroxide. Methods The superficies of cerebral cortexes were douched by 3% hydrogen peroxide in rats. The expression of glucocorticoiod receptor(GR), content of MDA and activty of SOD were determined in the douched brain tissues. The plasma level of glucocorticoid was detected. The pathologic changes in the douched brain tissues were observed. Results There were significant diffeences in MDA content, SOD activity and GR expression in the brain tissues, and plasma level of glucocorticoiod between the experimental group and control group (P<0.05). The significant damage to neurons, glial cells and vascular endotheliocytes was found out in the brain tissues douched with hydrogen peroxide. Conclusions Douching the brain tissues by hydrogen peroxide may produce fervent lipid peroxidation and stress reaction in the brain tissues, and damage to the brain tissues, in which the cerebrovascular injuiry may play an important role. It is suggested that douching the brain tissues by hydrogen peroxide should be cautiously performed during neurosurgical operation.
