小叶黑柴胡茎叶总黄酮对小鼠免疫性肝损伤的影响

摘    要:目的研究小叶黑柴胡茎叶总黄酮(TFA)对免疫功能的保护作用。方法采用碳粒廓清法比较正常组与小叶黑柴胡给药组的碳粒廓清指数;尾静脉注射刀豆蛋白(ConA,20 mg.kg-1)诱发小鼠免疫肝损伤模型,观察血清ALT和AST,并测定小鼠肝组织中MDA的含量。在光学显微镜下观察肝脏病理组织的变化。结果与正常组相比,TFA高、中、低(300、150、75 mg.kg-1)3个剂量组均能提高廓清指数K值(P<0.05)和校正廓清指数α值(P<0.01);能降低小鼠免疫肝损伤模型的ALT和AST活力和肝组织中MDA的含量。TFA能明显改善肝组织的病理学改变。结论 TFA能提高小鼠单核巨噬细胞系统的吞噬功能,使小鼠的免疫功能提高,对免疫性肝损伤小鼠有一定的保护作用。

Abstract:Objective To determine the effect of total flavonoids of the aerial part of Bupleurum smithii Wolff var.parvifolium(TFA) on immuno hepatic injury in mice.Methods Carbon expurgation method was used to compare the carbon expurgatory function of the control group and TFA groups in mice.ConA(Concanavalin A,20 mg·kg-1) in NS was injected intraperitoneally to establish an immunized mouse model of liver damage.We observed the serum level of ALT and AST.The liver tissue MDA,and the hepatic histological changes were observed by opticalmicroscope.Results Compared with that of the control group,the expurgatory index and phagocytic index increased(P〈0.05) in the 3 TFA groups(300,150,and 75 mg·kg-1).In the immunized models,the ALT and AST activity significantly decreased(P〈0.01),MDA content in the liver was also lowered(P〈0.01).TFA improved the hepatic pathological changes distinctly.Conclusion The total flavonoids of the aerial part of Bupleurum smithii Wolff var.parvifolium may protect immuno hepatic injury in mice.
