褪黑素对大鼠急性肾功能衰竭的保护作用及增强肝细胞生长因子的表达

摘    要:目的探讨褪黑索(Mel)在甘油诱导的急性肾功能衰竭(ARF)大鼠模型中的作用及对肾组织局部肝细胞生长因子(HGF)及其受体(C—met)表达的影响。方法将SD雄性大鼠随机分为3组:对照组(CTL组)双侧后腿肌肉注射生理盐水;模型组(ARF组)双侧后腿肌肉注射甘油;褪黑素组(Mel组)腹腔注射Mel后30min,双侧后腿肌肉注射甘油。分别在1、6、12、24、36、48、72、120h检测血尿素氮(BUN)、肌酐(Cr)、肾组织匀浆超氧化物歧化酶(SOD)、丙二醛(MDA)水平。观察肾组织形态学改变,并用免疫组织化学法检测HGF及C—met蛋白在肾脏的表达。结果与ARF组相比,Mel组肾功能(BUN、Cr)明显改善,肾组织匀浆SOD活性升高、MDA下降,肾小管上皮细胞HGF/C-met表达增强,差异均有显著性意义(P〈0.05或P〈0.01)。结论褪黑素对ARF大鼠肾脏有保护作用,能够增强HGF及其受体C—met蛋白的表达,促进损伤的肾小管恢复。

Abstract:Objective To investigate the effect of melatonin (Mel) on the prevention of experimental acute renal failure (ARF) induced by glycerol and observe the expression of hepatocyte growth factor (HGF) and recepator (C-met) in the rat kidney.Methods The SD rats were randomly divided into three groups: In group CTL, saline was intramuscularly injected into both thighs; In group ARF, glycerol was injected; In group Mel, melatonin plus glycerol was injected. The animals were sacrificed at 1st, 6th, 12th, 24th, 36th,48th, 72nd and 120th h after injection. Blood urea nitrogen, serum creatinine, superoxide diamutase (SOD) and malondiadehyde (MDA) were measured. The renal histopathological changes were observed under a light microscope. The expression of HGF and C-met was detected by immunohistochemistry.Results Compared with group ARF, mean blood urea nitrogen and serum creatinine concentrations were significatly decreased in group Mel, the activity of SOD was markedly increased and the level of MDA was decreased, and the expression of HGF and C-met protein was markedly increased in the renal tubular epithelial cells (P<0.05, P<0.01).Conclusion Melatonin can prevent the kidney of the rats with ARF, enhance the expression of HGF and C-met in renal tissue and promote recovery of the damaged renal tubular epithelial cells.
