陈皮、半夏对动脉粥样硬化小鼠PI3K-Akt通路、SOD、MDA、SA-β-gal水平的影响

摘    要:目的 探讨陈皮、半夏治疗动脉粥样硬化的机制。方法 将40只Klotho 敲除小鼠采用数字表法随机分为模型组、治疗组(高、中、低剂量组,每组10只)。对照组C57BL/6小鼠10只。通过HE染色观察小鼠颈动脉病理改变;通过血浆分光光度法检测丙二醛(malonaldehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、β-半乳糖苷酶(senescence-associated beta-galactosidase,SA-β-gal);通过免疫组化法观察PI3K与p-Akt表达。结果 与模型组比较,陈皮、半夏组动脉内膜呈轻度增生改变;SOD浓度与对照组比较上升,差异有统计学意义(P<0.01),MDA和SA-β-gal则显著降低(P<0.01);大剂量和中剂量治疗组中PI3K下降水平与模型组比较,差异有统计学意义(P<0.01);陈皮、半夏各治疗组p-Akt表达均显著低于模型组,差异有统计学意义(P<0.01)。结论 陈皮、半夏可能通过PI3K和p-Akt通路而调控SOD、MDA及SA-β-gal水平,达到延缓衰老、治疗动脉粥样硬化的作用。

Effects of citrus and pinellia on the levels of PI3K-Akt pathway,SOD, MDA,SA-sap-gal in atherosclerosis mice

Abstract:Objective To explore the mechanism of treating atherosclerosis with Citrus and Pinellia. Methods A total of 50 Klotho knockout mice were randomly divided into the normal group, the model group and the treatment group (high, medium and low dose group). The pathological changes of carotid artery were observed with HE staining. The plasma of malondialdehyde(mDA), superoxide dismutase(SOD), senescence-associated beta-galactosidas (SA-β-gal) and the expression of PI3K and p-Akt was examined with immunohistochemistry. Results Compared with the model group, the arterial intima in the Citrus and Pinellia group showed mild hyperplasia. Compared with the control group, the level of SOD increased significantly (P<0.01), while the content of MDA and SA-β-gal decreased significantly (P<0.01). Compared with the model group, the decrease level of PI3K in the high and medium-dose treatment groups was statistically significant (P<0.01), P-Akt expression in the treated group was significantly lower than that in the model group, and the difference was statistically significant (P<0.01). Conclusion Citrus and Pinellia may regulate the levels of SOD, MDA and SA-β-gal through PI3K and p-Akt pathway, to delay senescence and treat atherosclerosis.
