白细胞介素-25与支气管哮喘小鼠发病的关系

摘    要:目的探讨IL-25与支气管哮喘发病的关系。方法将30只健康雌性Balb/c小鼠随机分为3组:分别为卵白蛋白(OVA)致敏并激发的哮喘模型组(A组),OVA)致敏、激发并予糖皮质激素治疗组(B组)及正常对照组(C组),每组10只。于末次激发后24h后各组小鼠摘眼球取血,ELISA法检测其血清IL-25、IFN-γ及IL-4水平。收集支气管肺泡灌洗液(BALF),行嗜酸性粒细胞(EOS)计数,HE染色观察各组肺组织病理改变。RT-PCR法检测各组肺组织IL-25mRNA和IL-4mRNA表达。结果 1、B组多数小鼠未见明显行为学异常反应,肺组织病理损害较A组明显减轻,BALF中EOS计数(126.2±10.8)×103/L亦显著低于A组(324.7±13.6)×103/L(P〈0.01)。2、A组小鼠血清IL-25、IL-4水平[(80.66±3.06)pg/ml,(124.7±4.13)pg/m]均高于C组[(46.25±1.90)pg/ml,(32.73±2.20)pg/ml],IFN-γ水平[(45.25±5.34)pg/ml]低于C组[(80.56±2.49)pg/ml]其差异均有统计学意义(P〈0.01)。3、A组小鼠肺组织IL-25mRNA和IL-4mRNA表达均高于C组(P〈0.01),B组小鼠肺组织IL-25mRNA和IL-4mRNA表达均低于A组(P〈0.01)。A组小鼠肺组织IL-25mRNA表达量与BALF中EOS计数呈正相关(r=0.901,P〈0.01)。结论 IL-25参与了哮喘小鼠的发病,在哮喘的发病中起促炎作用,其促进哮喘发病的作用可以被激素抑制。

Abstract:Objective TO explore the role of IL-25on bronchial asthma.Methods Thirty female BALB/c mice were randomly divided into 3group(n=10):asthma model(groupA),corticosteroid treatment group(groupB)and normal control group(groupC).All mice were killed 24hafter final OVA challenge.Blood was obtained for measurements of aerum IL-17,TFN-γand IL-23by ELISA.Bronchoalveolar lavage fluid(BALF)was collected for eosinophil(EOS) count.The left lung was isolated for pathological examination.Lung sections were stained with hematoxylin and eosin(HE).The mRNA expressions of IL-25in lung tissue were measured by RT-PCR.Results The mouse asthma model appeared ethology change specific to ashma,EOS count increase in BALF and typical pathological change in lung tissue.The BALF EOS count[(324.7±13.6)×103/L],the concentrations of serum IL-25[(80.66±3.06)pg/ml,(124.7± 4.13)pg/ml]and mRNA expressions of IL-25in group A were significantly higher than those in group C,while the concentration of serum TFN-×[(45.25±5.34)pg/ml]IN Group A was significantly lower than that in group C(P 0.01).The mRNA expressions of IL-25were significantly lower than those in group A,while the concentration of serum TFN-γ[(80.56±2.49)pg/ml]was significantly higher than that in group A(P0.01).Conclusion The IL-25 levels with increased concentrations in serum and mRNA expression in asthmatic mouse lung might be involved in the pathogenesis of bronchial asthma,and could be inhabited by inhaled glucocorticoid.
