摘    要:目的 观察大黄素对小鼠动脉粥样硬化(AS)模型血脂、血清炎性因子等的影响,评价大黄素在降脂和抗动脉粥样硬化方面的作用.方法 将60只雄性apoE-/-小鼠适应性饲养1周后,随机分为模型组(n=15)和大黄素组(n=45),大黄素组又随机分为高剂量组(1200 mg/kg·d)、中剂量组(900 mg/kg·d)和低剂量组(600 mg/kg·d)三个亚组,每组15只.正常对照组给予普通颗粒饲料;模型组给予高胆固醇饲料(0.25%胆固醇+15%脂肪),大黄素组给予高胆固醇饲料+大黄素,喂养12周处死.处死前动物称量体重、血脂检测、血清炎性因子检测.结果 12周末模型组小鼠体重增加显著高于大黄素高剂量组(P<0.05);大黄素治疗组TC水平均显著降低(P<0.05~0.01),且药物的作用呈现明显的剂量依赖性(P<0.05);三个大黄素治疗组hsCRP水平和Fib浓度均显著降低(P<0.05~0.01),且中剂量组和低剂量组均显著高于高剂量组.结论 大黄素对AS的发生具有全面的干预作用.

Abstract:Objective: To observe emodin atherosclerosis(AS) model of serum blood fat in mice and the effect of inflammatory factor,and to evaluate emodin in lipid-lowering and resistance of the role of atherosclerosis.Methods:Sixty males apoE-/-mice fed after one week were randomly divided into the model group(n = 15) and the emodin group(n = 45),and then the emodin group were randomly divided into the high dose group(1200 mg/kg·d),the dosage group(900 mg/kg·d.) and the low dose group(600 mg/kg·d),with 15 mice in every group.The normal control group were given ordinary particles fodder,the model group were given high cholesterol feed(0.25% cholesterol + 15% fat),and the emodin group were given high cholesterol + emodin.All the mice were put to death after 12 weeks feeding.All the animals were weighed and blood fat and serum inflammatory factor were detected before being put to death.Results: The weight of emodin mice model was significantly higher than that of the high dose group(P<0.05) at 12 weekend,and the emodin treatment group had significantly lower level of TC(P<0.05 ~ 0.01),and the effect of medicines was obviously dose dependent(P< 0.05).The hs CRP level in all the three emodin treatment groups had significantly lower Fib concentration(P<0.05 ~ 0.01),and those of low dose group and dosage group were significantly higher than that of the high dose group.Conclusion: Emodin has an overall role of intervention with the occurrence of AS.
