| 摘 要： | 目的探讨硫化镍诱导人支气管上皮(16HBE)细胞恶性转化作用以及扶正解毒汤(FJD)的保护作用,研究硫化镍致癌的分子机制以及扶正解毒汤对防癌治癌的作用。方法用不同浓度的硫化镍在体外多次处理16HBE细胞,利用刀豆凝集素A(ConA)凝集实验和软琼脂集落形成实验进行转化细胞的恶性鉴定;测定16HBE细胞内镍离子和自由基含量,同时加入扶正解毒含药血清,观察其保护作用。结果硫化镍可诱导16HBE细胞的恶性转化,转化细胞的生长速度加快、排列紊乱、失去接触抑制、呈交叉重叠生长、转化率呈剂量—效应关系。转化细胞可被低浓度ConA凝集并在软琼脂内生长。硫化镍暴露组细胞内镍离子和自由基含量明显增加。中、高剂量的扶正解毒含药血清可通过细胞内、外途径发挥抗氧化、清除自由基效应。结论硫化镍有较强的诱导16HBE细胞恶性转化的能力,具有致癌性。FJD血清对于硫化镍诱导的16HBE细胞恶性转化具有保护作用。
