补骨脂抗氧化及抗肿瘤活性成分的研究

目的：研究补骨脂成分的抗氧化和抗肿瘤活性。方法：运用多种柱色谱等现代分离手段对补骨脂甲醇浸膏进行分离,利用磁共振技术和电喷雾电离质谱鉴定化合物的结构,并评价化合物的抗氧化和抗肿瘤活性。结果：分离出11个化合物,鉴定为：①补骨脂素,②异补骨脂素,③补骨脂定,④补骨脂苷,⑤异补骨脂苷,⑥双补骨脂酚A,⑦双补骨脂酚B,⑧12,13-二氢-12,13-环氧补骨树脂酚；⑨补骨脂宁,⑩异补骨脂二氢黄酮,?补骨脂二氢黄酮甲醚。化合物③,⑥,⑦具有清除1,1-二苯基-2-三硝基苯肼自由基的能力；③,⑨~?对肝癌HepG2细胞和乳腺癌MDA-MB-231细胞具有抗增殖作用,⑦对肝癌HepG2细胞有一定抗增殖作用。结论：化合物③,⑥,⑦具有一定的抗氧化活性；③,⑦,⑨~?具有一定的抗肿瘤活性。

Objective:To screen the anti-oxidative and antitumor compounds from Psoralea corylifolia L. seeds. Methods:The methanol extract of Psoralea corylifolia L. was separated by a variety of column chromatography and the modern separation methods,the compounds were identified by nuclear magnetic resonance and electrospray ionization-mass spectrum. Results:Eleven compounds were obtained and identified as ① psoralen,② isopsoralen,③ psoralidin,④ psoralenoside,⑤ isopsoralenoside;⑥ bisbakuchiols A,⑦bisbakuchiols B,⑧ 12,13- dihydro- 12,⑨13-epoxybabakuchiol;corylin,⑩ isobavachin,? and bavachinin. Compounds ③,⑥,⑦shown scavenging effect to the 1,1-Diphenyl-2-picrylhydrazyl radical;compounds ③,⑨-? displayed the antiproliferation effect to the liver cancer HepG2 and breast cancer MDA-MB-231 cells,compounds⑦displayed the antiproliferation effect to the liver cancer HepG2 cells. Conclusions:Compounds ③,⑥,⑦ shown antioxidant activity;compounds ③,⑦,⑨ -? shown antitumor activity.
