摘    要:目的 探讨长链非编码RNA核富集转录本1(lncRNA NEAT1)在精神分裂症(sch)病人外周血中的表达与临床症状的相关性。方法 选择2019年6月至2022年3月在菏泽市第三人民医院及河北医科大学第一医院精神科收治的sch病人73例作为观察对象(sch组),另选取同期在该两家医院体检人员(无精神病障碍史及无精神疾病家族史)68例作为对照组,采用阳性与阴性症状量表(PANSS)评估病人临床症状,采用蒙特利尔认知评估量表(MoCA)评估病人认知功能,实时荧光定量逆转录聚合酶链反应(qRT-PCR)法检测外周血单核细胞(PBMC)中lncRNA NEAT1表达水平,Spearman法分析lncRNA NEAT1与PANSS评分、MoCA评分的相关性,受试者操作特征(ROC)曲线分析lncRNA NEAT1水平对sch的诊断价值。结果 对照组和sch组病人外周血lncRNA NEAT1表达水平分别为1.05±0.21、0.73±0.15,差异有统计学意义(P<0.001);ROC曲线分析显示:lncRNA NEAT1的曲线下面积为0.82,截断值为0.89,灵敏度为79.45%,特...

Abstract:Objective To investigate the correlation between the expression of long noncoding RNA nuclear enriched transcript 1(lncRNA NEAT1) in the peripheral blood of patients with schizophrenia (sch) and clinical symptoms.Methods A total of 73 sch pa.tients admitted to the Psychiatry Department of the Heze Third People''s Hospital and the First Hospital of Hebei Medical Universityfrom June 2019 to March 2022 were selected as the observation subjects (sch group), and the other 68 patients who received physicalexaminations in these two hospitals during the same period (without history of mental disorders and no family history of mental disor.ders) were chosen as the control group. The patients were assessed for their clinical symptoms by using the Positive and Negative Syn.drome Scale (PANSS). The Montreal Cognitive Assessment Scale (MoCA) was used to assess the cognitive function of the patients, andthe expression level of lncRNA NEAT1 in peripheral blood mononuclear cells (PBMCs) was detected by real-time fluorescence quanti. tative reverse transcription-polymerase chain reaction (qRT .PCR) assay. The correlation between the lncRNA NEAT1 and PANSSscores and MoCA scores was analyzed by Spearman''s method, and the diagnostic value of sch was analyzed by receiver operating char.acteristic (ROC) curves for the level of lncRNA NEAT1.Results The expression levels of peripheral blood lncRNA NEAT1 in pa.tients in the control group and sch group were 1.05±0.21 and 0.73±0.15, respectively, and the difference was statistically significant (P< 0.001). ROC curve analysis showed that the area under the curve of lncRNA NEAT1 was 0.82, and the cutoff value was 0.89, with asensitivity of 79.45% and a specificity of 73.93%. Correlation analysis showed that the lncRNA NEAT1 was negatively correlated withthe total score of the PANSS scale and the score of the Negative Symptoms Scale (r=.0.40, .0.49, P<0.05) and was negatively correlat. ed with executive ability, attention, delayed memory, and the total score of the MoCA scale (r=.0.38, .0.40, .0.40, .0.42, P<0.05). Conclusion The reduced expression level of lncRNA NEAT1 in the peripheral blood of sch patients is associated with the mental symptoms and cognitive function of sch patients and may be involved in the occurrence and development of sch.
