电针对大鼠胃黏膜损伤相关信号分子的影响

摘    要:目的 探索电针对胃黏膜损伤大鼠胃黏膜细胞损伤修复的作用机制.方法 将大鼠随机分为正常组、模型组、针刺治疗组,每组10只,应用乙醇灌胃法造成胃黏膜损伤大鼠模型,用电针对胃黏膜损伤大鼠治疗10天,采用链霉蛋白酶消化法分离胃黏膜细胞,用表面加强激光解析电离飞行时间质谱技术(SELDI-TOF-MS)及WCX2蛋白芯片获得各组大鼠胃黏膜细胞的蛋白质指纹图谱,对比分析各组的差异蛋白质质/荷比峰.结果 模型组大鼠胃黏膜细胞蛋白质SELDI-TOF-MS蛋白指纹图谱与正常组比较有4个蛋白质质/荷比峰差异有统计学意义(P<0.05或P<0.01),其中两个蛋白质质/荷比峰明显升高(P<0.05或P<0.01),两个蛋白质质/荷比峰明显降低(P<0.05);针刺治疗组与正常组比较共有4个蛋白质质/荷比峰差异有统计学意义(P<0.05或P<0.01),其中1个蛋白质质/荷比峰明显升高(P<0.05),3个蛋白质质/荷比峰明显降低(P<0.05或P<0.01);针刺治疗组与正常组比较亦有4个蛋白质质/荷比峰差异有统计学意义(P<0.05或P<0.01),且4个蛋白质质/荷比均为降低(P<0.05或P<0.01).结论 电针对胃黏膜损伤的修复可能不是单纯针地对某一个或两个蛋白的调节发挥治疗作用的,而可能是多个蛋白质参与的复杂的链环反应.

Effect of Electroacupuncture on Gastric Mucosal Damage Related Signal Molecules in Rats
Abstract:Objective To study the repairing mechanism of ekctroacupuncture on mucosal cells of rats with gastric mucosal damage.Methods The experimental rats were randomized into normal group,model group,and electroacupuncture group,10 in each.The models were created by gastric administration with ethanol.After electroacupuncture treatment for 1 0 days,the gastric mucosal cells were separated with pronase digestion method.The surface enhanced laser desorption ionization-time of flight-mass spectrometry (SEI-DI-TOF-MS)technique and WCX2 protein chip were applied to obtain the fingerprints of proteins of gastric mucosal cells of all groups.A contrastive analysis was carried out to see the difference in charge-to-mass ratio peaks of all groups.Resul ts Compared with the normal group,the fingerprints of proteins of gastric mucosal cells in the model group had 4 charge-to-mass ratio peaks,the difference was significant(P<0.05 or P<0.01),of which,two of the peaks were significantly elevated(P<0.05 or P<0.01),and two significantly lowered(P<0.05).Compared with the model group,the fingerprints of proteins of gastric mucosal cells in the electroacupuncture group had 4 charge-to-mass ratio peaks,the difference was significant(P<0.05 or P<0.01),of which,one of the peaks was significantly elevated(P<0.05),and three significantly lowered(P<0.05 or P<0.01).Compared with the normal group,the fingerprints of proteins of gastric mucosal cells in the electroacupuncture group also had 4 charge-to-mass ratio peaks,the difference was significant(P<0.05 or P<0.01),all of which were lowered(P<0.05 or P<0.01).Conelusion The repairing of gastric mucosal cells by electroacupuncture is probably not aimed at regulating one or two proteins,but a complicated chain reaction joined by multiple proteins.
