益脑康对动脉粥样硬化性急性脑缺血大鼠血清NO、VEGF及PDGF-BB含量的影响

摘    要:目的:观察中药益脑康胶囊对动脉粥样硬化(AS)基础上急性脑缺血(ACI)SD大鼠血清一氧化氮(NO)、血管内皮生长因子(VEGF)、血小板衍化生长因子-BB(PDGF-BB)含量的影响,以探讨益脑康胶囊防治缺血性中风的可能机制。方法:将体质量(180±20)g SD雄性大鼠随机分为A、B、C、D、E、F(即正常对照组、益脑康预防组、AS模型组、AS+ACI模型组、益脑康治疗组和立普妥治疗组)6组,采用腹腔注射VD3+高脂喂养复制AS模型,内皮素-1在大脑中动脉附近注射法复制ACI模型。B组在造模同时给予益脑康灌胃治疗,其余各组大鼠在模型成功后分别予0.9%氯化钠溶液、益脑康和立普妥灌胃治疗1周,在治疗前后分别测定血清NO、VEGF、PDGF-BB的含量。结果:B组、F组大鼠血清NO含量较D组明显增高(P0.01,P0.05)。B组、E组、F组与D组比较,血清VEGF、PDGF-BB水平均明显下降(P0.01,P0.05)。结论:益脑康胶囊可通过提高血清NO含量,降低VEGF、PDGF-BB含量发挥防治动脉粥样硬化性急性缺血中风的作用。

Abstract:Objective:By observing the effects of Yinaokang Capsule on NO,VEGF and PDGF-BB content in serum of atherosclerotic acute cerebral ischemic SD rats,the possible mechanism of the capsule for preventing ischemic stroke is investigated.Methods:SD male rats weighted(180±20)g were randomly divided into 6 groups:A,B,C,D,E,F(Respectively for the normal control group,the Yinaokang Capsule prevention group,the AS model group,the AS + ACI model group,the Yinaokang Capsule treatment group and the Lipitor treatment group).AS model is replicated by intraperitoneal injection of VD3 + high fat diet,and ACI model is replicated by the injection of ET-1 near the MCA.Rats of group B were treated with Yinaokang gastric perfusion while the modeling,the other rats were given saline,Yinaokang and Lipitor by gavage 1 week after successful modeling.The levels of NO,VEGF,and PDGF-BB in serum were measured before and after treatment.Results:NO content in serum of group B and group F was significantly higher than that of group D(P<0.01,P<0.05).Compared with group D,VEGF and PDGF-BB levels in the serum of Group B,E,F were significantly decreased(P<0.01,P<0.05).Conclusion:By increasing serum NO content and decreasing the content of VEGF,PDGF-BB,Yinaokang Capsule plays the role of prevention and treatment in rats of atherosclerotic acute ischemic stroke.
