摘    要:目的 :探讨新型生物降解材料聚已内酯 ( PCL ) /聚乳酸 ( PLA)共聚物与胎体雪旺氏细胞( FSCs)制备人工周围神经模型的可行性。方法 以 PCL /PLA共聚物为原料 ,模拟制备周围神经细胞外基质框架结构。将体外分离、培养的 FSCs种植于 PCL/PLA框架结构内。通过观察细胞生长状况及反转录 PCR半定量检测胶质细胞源神经营养因子 ( GDNF)基因表达情况 ,分析其相容性及促神经再生潜能。结果 FSCs能够良好地生长于 PCL/PLA框架结构内 ,PCL/PLA对 FSCs的GDNF基因表达无明显影响。结论 PCL/PLA共聚物与 FSCs具有良好的相容性及促神经再生潜能 ,可用于修复周围神经缺损的实验研究

Abstract:Objective The purpose of this study is to explore the feasibility of using the biodegradable polycaprolactone(PCL)/polylactide(PLA) copolymer and the fetal Schwann cells(FSCs) to develop the artificial peripheral nerve model of tissue engineering.Methods The regeneration tube frame was made with PCL/PLA copolymer. It consists of both double layered tube with rough interior and smooth exterior and inside W-shaped frame with rough surface membrane. The FSCs were cultured and isolated in vitro, then seeded in the nerve regenerative tube frame. Five days later, the growth condition of the FSCs was observed and the change in GDNF gene expression was analysed by semi-quantitative RT-PCR.Results It was found that a large number of FSCs had adhered to the surface of inside frame and the cells grew well. The PCL/PLA copolymer did not affect the function of GDNF gene expression of FSCs.Conclusion There is a good compatibility between PCL/PLA copolymer and FSCs which enables the FSCs to carry out their normal function in promoting nerve regeneration.
