乳腺癌中c-erbB-2癌基因和mdr-1基因的检测及相关性研究

 目的　检测原发性乳癌中c erbB 2扩增和mdr 1表达的情况 ,并探讨二者的相关性。方法　分别采用PCR和RT PCR法检测 47例乳癌中c erbB 2扩增和mdr 1表达。结果　⑴c erbB 2扩增与组织学分级和腋淋巴结阳性数有关 ,有统计学意义。ER、PR阴性与阳性乳癌比较 ,非浸润性与浸润性乳癌比较 ,c erbB 2扩增均有显著性差异。⑵mdr 1表达与组织学分级有关 ,有统计学意义。⑶mdr 1表达与c erbB 2扩增之间无统计学意义。结论　c erbB 2扩增与乳癌的组织学分级、腋淋巴结阳性数、ER、PR状态、组织学类型有关 ,mdr 1表达只与组织学分级有关 ,c erbB 2扩增与mdr 1表达无关

Study on the correlation between c -erbB -2 amplification and mdr -1 expression in breast cancer

Objective To investigate the c\|erbB\|2 amplification and the mdr\|1 expression in primary breast cancer and to reveal the correlation between these two genes.Methods PCR and RT\|PCR was used respectively to examine the c\|erbB\|2 amplification and mdr\|1 expression in 47 primary breast cancer. Results (1) The c\|erbB\|2 amplification was significantly linked to histological grade and the axillary lymph node involvement. Statistically significant difference in c\|erbB\|2 amplification was observed not only between ER/PR\|negative and ER/PR\|positive tumors , but also between non\|infiltrating and infiltrating tumors.(2)mdr\|1 expression was strongly correlated with histological grade.(3) No significant corrlation between the amplification of c\|erbB\|2 and the expression of mdr\|1 was found. Conclusion c\|erbB\|2 amplification was significantly associated with histological grade, axillary lymph node involvement, ER,PR status and histological subtype. Only histological grade was associated with the expression of mdr\|1.No significant correlation was observed between c\|erbB\|2 amplification and mdr\|1 expression.
