黄芪注射液诱导人结肠癌Lovo细胞凋亡机制的研究

目的研究黄芪注射液诱导结肠癌Lovo细胞株的凋亡机制。方法以不同浓度的黄芪注射液作用于人结肠癌Lovo细胞株,用MTT法测黄芪注射液对Lovo细胞体外增殖的影响,透射电镜观察细胞超微结构;流式细胞仪AnnexinV和PI双重染色行凋亡相关检查。结果与对照组相比,实验组对Lovo细胞株增殖均有明显的抑制作用,且与浓度和作用时间呈正比;电镜下可见凋亡的早、中、晚期细胞及凋亡小体,同时可见部分细胞呈坏死改变。流式细胞仪显示:黄芪注射液作用Lovo细胞48 h后,0.05 g/mL、0.1 g/mL、0.15 g/mL、0.2g/mL各组的凋亡率分别为4.8%,18.6%,42.1%,64.8%。结论黄芪注射液能抑制Lovo细胞生长,诱导细胞凋亡。

Mechanism study of Huangqi injection inducing to apoptosis of human colon cancer Lovo cell

Objective It is to study the mechanism of Huangqi injection inducing to apoptosis of colon cancer Lovo cell lines.Methods Different concentrations of Huangqi injection were applied on human colon cancer Lovo cell lines.The influence of Huangqi injection on proliferation of Lovo cell in vitro was detected with MTT method.The ultrastructure of Lovo cell was observed with electron microscopy.The detections related apoptosis were detected with flow cytometer AnnexinV and PI double staining.Results Compared with control group,the proliferation of Lovo cell lines in experimental group had been obviously inhibited,and the relation with the concentration and the action time was direct proportion.The cell apoptosis in early,middle and advanced stages and apoptotic body were seen under electron microscope,necrosis change was seen in some cells at the same time.FCM showed that the apoptosis rates in groups of 0.05 g/mL,0.1 g/mL,0.15 g/mL and 0.2 g/mL were 4.8%,18.6%,42.1% and 64.8% respectively after Huangqi injection on Lovo cell for 48 h.Conclusion Huangqi injection can inhibit the growth of Lovo cell and induce cell apoptosis.
