川芎嗪对脑缺血再灌注后神经细胞凋亡及其相关蛋白Fas-L表达的影响

摘    要:目的观察川芎嗪对大鼠局灶性脑缺血再灌注后神经细胞凋亡及Fas-L蛋白表达的影响。方法SD大鼠36只随机分成假手术组、模型组、川芎嗪组。采用TUNEL法检测各组大鼠脑组织神经细胞凋亡;采用免疫组织化学法与医学图像分析结合的方法检测各组大鼠脑组织Fas-L蛋白的表达。结果与假手术组相比,模型组大鼠缺血侧脑组织TUNEL阳性神经细胞及Fas-L阳性细胞表达增多;与模型组相比,川芎嗪组大鼠缺血侧脑组织TUNEL阳性神经细胞及Fas-L阳性细胞表达减少。结论川芎嗪可下调Fas-L蛋白的表达,从而抑制脑缺血再灌注后神经细胞凋亡,这可能是其对脑缺血再灌注损伤起保护作用的分子机制之一。

Abstract:Objective: To investigate the effects of ligustrazine on apoptosis of neuronal cell and on the expression of apoptosis-associated genes Fas-L proteins after cerebral ischemia and reperfusion in rats.Methods: 36 SD rats were divided randomly into the sham-operated group, the model group and the ligustrazine gruop. Apoptosis of neuronal cell in every group rats'brain tissues were analyzed by TdT-mediated dUTP-biotin nick end labeling (TUNEL). The immunohistochemistry and MIPS were used to measure the expression of Fas-L positive cells in rats' cerebral cortex of every group.Results: Compared wiht those in the sham-operated group, the expression of postitve neuronal cells of TUNEL and positive cells of Fas-L protein increased in model group rats' ischemic brain tissues. Compared with those in the model group, the expression of positive neuronal cells of TUNEL and Fas-L protein decreased in ligustrazine group.Conclusion: Ligustrazine could inhibit neuronal cell apoptosis by down-regulating the expression of Fas-L protein, which may be one of mechanisms of neuroprotective effects of ligustrazine.
