多形核粒细胞质膜结合酶活性的氧化损伤及IH-764-3的抗损伤作用

摘    要:用乙酸肉豆寇咐醇酯(PMA)刺激人多形核粒细胞(PMN)可导致其质膜结合酶活性下降,碱性磷酸酶(ALP)、5'-书苷酸酶(5'-NT)和中性内肽酶(NEP)活性分别降至36.61%、58.53%和30.79%。及H_2O_2生成抑制试验证明,PMA刺激后PMN质膜结合酶活性的损伤乃活性氧物质(ROI)所致。IH-764-3是自中药中提取的一种单体成分,经实验证明具有抗氧化损伤,保护PMN质膜结合酶的作用,该抗损伤作用可能与其抑制H_2O_2生成有关。

Abstract:olymorphonuelear leukocytes(PMN)stimulatedwith phorbol myristate acetate (PMA) induced the de-creases of plasma membrane-bound enzyme activities ofPMN themselves.The activities of alkaline phosphatase(ALP),5'-nucleotidase (5'-NT) and neutral endopepti-dase(NEP)decreased to 36.61 %,58.53% and30. 79%,respectively.The test of and H_2O_2 pro-duction supported that the damage of the plasma mem-brane-enzymes of PMN was caused by reactive oxygenintermediates (ROI).IH-764-3, an active monomercomponent from Chinese herbs, showed the protectiveeffect to plasma membrane-bound enzymes against theoxidative damage,which was related to inhibit the gen-eration of H_2,O_2 by activated PMN themselves.
