白芍总苷对HepG2细胞增殖的抑制作用

目的 探讨白芍总苷（TGP）对人肝癌细胞株HepG2的增殖抑制作用。方法 采用Mtt法检测TGP对HepG2细胞增殖的影响；应用荧光显微镜观察药物作用后的细胞形态；用流式细胞术检测TGP对细胞凋亡及细胞周期的影响。结果 TGP（1．0～5．0g／L）能抑制HepG2细胞生长，且呈浓度依赖性；TGP（1．5、2．5g／L）分别作用72h后，HepG2细胞出现体积缩小，荧光染色增强，胞核或胞质中可见致密浓染的块状或颗粒状黄绿色荧光染色；TGP（1．0、1．5g／L）药物作用72h后，流式细胞仪可检测到细胞凋亡现象并发现显著的S期阻滞。结论 TGP在体外能够抑制人肝癌细胞HepG2的增殖，并能诱导细胞凋亡。

Proliferation inhibition and mechanisn of TGP on HepG2 Cell in vitro

Objective To study the proliferation inhibition of total glucosides of paeony(TGP) on human hepatoma cell HepG2. Methods The proliferation inhibitory effect of TGP on tumor cells was assayed using MTT method. The morphologic changes of the human hepatoma cell HepG2 were observed via staining with acridine orange (AO)fluorescent dyes. The effect of TGP on cell-cycle and apoptosis was analyzed using flow cytometry(FCM). Results TGP (1.0~5.0 g/L) had inhibitory effect on the proliferation of HepG2 concentration-dependently. AO dying showed TGP induced typical chromatin condensation in HepG2 cells. The result of FCM showed HepG2 cell apoptosis after treated with TGP for 72 h and the cell cycle was arrested at S phase. Conclusion TGP can inhibit the proliferation of HepG2 cells in vitro and induce apoptosis.
