Protective effect of ethyl pyruvate on renal injury in severe acute pancreatitis rats

Abstract:Objective To explore the therapeutic role and its mechanism of ethyl pyruvate on renal injury induced by severe acute pancreatitis(SAP) in rats.Methods Seventy-two SD rats were randomly divided into three groups: sham operation group(S group),SAP group(P group) and ethyl pyruvate treatment group(T group).SAP model was established by retrograde infusion of 50 g/L sodium taurocholate into the biliopancreatic duct of rats.The rats were killed at 3 h,6 h and 12 h after operation.The serum level of TNF-α,IL-6,BUN,Cr and HMGB-1 were measured.The expression of NF-κB was detected by immunohistochemistry.The renal tissues were taken for HE staining and electron microscopic examination.Results In group P,serum levels of TNF-α,IL-6,BUN,Cr and expression of NF-κB in the renal tissues were higher than those in group S(P<0.05),HMGB-1 levels were higher than those in group S at 6 h and 12 h(P<0.05).The serum level of TNF-α,IL-6,BUN,Cr,HMGB-1 and expression of NF-κB in the renal tissues in T group were lower than those in group P(P<0.05).Renal pathological damage was milder in group T than those in group P under microscopy and electron microscopy.Conclusion Ethyl pyruvate plays a protective role against renal injury in severe acute pancreatitis rats.The underlying mechanism may be related to inhibition of TNF-α,IL-6,HMGB-1 and suppression of NF-κB.
