实验性颈动脉粥样硬化纤溶系统活性与增殖细胞核抗原表达的观察

摘    要:目的:观察在用空气干燥加高脂饮食法建立颈动脉粥样硬化(AS)模型过程中纤溶系统活性与增殖细胞核抗原(PCNA)表达的变化。方法:54只日本大耳兔分为3组,分别给予空气干燥加高脂饮食(n=24)、单纯高脂饮食对照(n=24)和正常饮食对照(n=6),在实施空气干燥后的第3天、1w、2w、4w分别处死动物,观察颈动脉内膜和中膜的PCNA表达,测定不同时点的纤溶系统活性。结果:空气干燥加高脂饮食组出现较典型的AS病变,2w、4w时内膜增生明显,内膜与中膜PCNA表达明显增高,第4周时抗凝血酶Ⅲ(AT-Ⅲ)明显降低而纤溶酶原(PLG)明显升高。结论:空气干燥加高脂饮食法可以形成典型的颈AS病变,病变形成与纤溶系统活性改变、内膜中膜细胞增生有关。

Abstract:Objective:To observe the changes of coagulation and fibrinolysis systems and cell proliferation activity in experimental carotid atherosclerosis model using air-drying in the cholesterol-fed rabbits.Methods:54 male Japanese White rabbits were randomly divided into 3 groups. Group 1 (n = 24) was fed by high-cholesterol diet and treated by air-drying as experimental group. Other two groupsas empty control (n= 6) and cholesterol-fed control group (n = 24). Animals were sacrificed at various times (from 3 days to 4 weeks) afte.r sundry in the experimental group and at the corresponding times in the control group. The cell proliferation activity in the intima and media were quantified by immunocytochemistry for PC-NA. At the same time, the level of the blood lipid^ AT- III and PLG were detected. Results:The typical carotid atherosclerosis was observed in the experimental groups. The AT- III and PLG level had significant changes in experimental group at the 4th week compared with other groups. The distribution of the the PCNA positive cells in media and intima also increased in experimental group at the 3rd and 4th weeks. Conclusion:This study shows that the distribution of the PCNA positive cells and the changes of coagulation and fibrinolysis systems were corresponded with the pathological changes.
