| 摘 要： | 目的：探讨嗜酸性粒细胞活化支气管上皮细胞后对其细胞因子白细胞介素（IL）-6分泌的影响。方法：正常和1％多聚甲醛固定的嗜酸性粒细胞分别与支气管上皮细胞（BEAS-2B）细胞联合培养，应用流式细胞微珠实验（CBA）方法定量细胞培养上清液中细胞因子的浓度，用RT-PCR方法分析经嗜酸性粒细胞活化后支气管上皮细胞中IL-6的基因表达。结果：经嗜酸性粒细胞活化后，支气管上皮细皮中IL-6基因表达显著上调，细胞培养上清液中IL-6蛋白质浓度升高，多聚甲醛固定后的嗜酸性粒细胞能够刺激支气管上皮细胞中IL-6基因表达和蛋白质释放。结论：嗜酸性粒细胞可活化支气管上皮细胞中IL-6基因表达和促进其蛋白质分泌。 |

| Abstract: | Objective:To investigate the effect of human epithelial cells activation by co-culture with human eosinophils on interleukin(IL)-6 expression.Methods:Normal and 1% paraformaldehyde fixed eosinophils were co-cultured with epithelial cells(BEAS-2B cells) respectively;IL-6 gene expression in BEAS-2B cells was evaluated by RT-PCR and IL-6 protein in culture supernatant was determined by flow cytometry with CBA kit.Results:The interaction of eosinophils and BEAS-2B cells was found to up-regulate IL-6 gene expression in BEAS-2B cells and increase IL-6 release in co-culture supernatant.Conclusion:Above results showed that eosinophils can activate epithelial cells to release IL-6. |
