HPLC-ELSD法测定复方苦参洗剂中苦参碱的含量

目的建立复方苦参洗剂中苦参碱的含量测定方法。方法采用高效液相-蒸发光散射检测法测定苦参碱的含量。Hypersil NH2色谱柱（250mm×4．6mm，5μm），乙腈旬．1％的三乙胺（68：32）为流动相，流速1．0ml·min^-1，柱温25℃。ELSD参数为：漂移管温度94．4℃，载气流速2．4L·min^-1。结果苦参碱质量（X）的对数值与色谱峰面积（Y）的对数值之间呈良好的线性关系，回归方程为lgY=0．908 lgX＋5．64，r=0．9999，线性范围为0．3456—5．184μg，检测限为0．0432μg，定量限为0．1296μg，平均回收率为98．36％（RSD为0．82％）。结论本法操作简便，结果准确，重现性好，可用于测定苦参碱的含量。

Determination of matrine in compound kushen lotion by HPLC-ELSD

Aim To establish a method to determine matrine in Compound Kushen lotion.Methods HPLC-ELSD was used to determine the matrine.Hypersil NH2 column(250 mm×4.6 mm,5 μm)was used as the chromatographic column,acetonitrile-0.1% triethylamine(68∶32)as mobile phase,with a flow rate of 1.0 ml·min-1,and column temperature of 25℃.The tube temperature was 94.4 ℃,and the nitrogen flow rate was 2.4 L·min-1.Results The calibration curve for matrine was linear over the range of 0.3456~5.184 μg(r=0.9969).The average recovery was 98.36% with RSD of 0.82%(n=9).The limit of detection for matrine was 0.0432 μg.The limit of quantification for matrine was 0.1296 μg.Conclusion The established method is simple,accurate and can be used to determine the matrine in compound kushen lotion.
