摘    要:目的建立测定黄芪破壁饮片中黄芪甲苷含量的高效液相-蒸发光散射检测的方法。方法采用C18柱(5μm,200mm×4.6mm),乙腈-水(32:68)为流动相,流速1.0mL/min,蒸发光散射检测器漂移管温度80%,载气流速2.7mL/min。结果该方法回收率99.5%(RSD=2.0%),精密度RSD=I.4%,重复性RSD=2.5%,黄芪甲苷在(1.2913-10.330)μg范围内线性关系良好(r=0.9998)。结论该方法简便快速,可作为黄芪破壁饮片中黄芪甲苷的含量测定方法。

Abstract:Objective To establish a method to determinate astragaloside content in astragalus broken-cell pieces by HPLC-ELSD. Methods The samples were separated on a C 18 (250 mm×4.6 mm, 5 μm)column with the mobile phase consisted of acetonitrile-water (32:68) at the flow rate of 1.0mL/min.The drift tube tempreture of the ELSD was 80℃ and the flow rate of carrier gas was 2.7 mL/min .Results The average recovery rate was 99.5% (RSD=2.0%).A good linearity range was shown at the concentration of (1.291-10.330)μg (r = 0.9998). Conclusion The method is simple practical and can be used for determination of astragaloside content in astragalus broken-cell pieces.
