大鼠甲状旁腺的切取及功能测定

摘    要:目的 通过对大鼠甲状旁腺的切取方法及功能的研究 ,为甲状旁腺细胞的移植奠定基础。方法 手术切取甲状旁腺 ,并行病理学检查。同时 ,用胶原酶和胰蛋白酶消化甲状旁腺 ,进行细胞培养。对培养5d的甲状旁腺细胞做透射电镜检查 ,应用放射免疫分析对 1d、3d、5d、7d的细胞培养液行甲状旁腺激素测定。结果  18只鼠的甲状旁腺被成功切除 ,切除率 90 % (18/ 2 0 )。培养 5d的甲状旁腺细胞具有丰富的粗面内质网、高尔基复合体、杆状的线粒体和分泌颗粒。培养的甲状旁腺细胞可以正常分泌甲状旁腺激素 ,以第 5d为最佳。结论 大鼠甲状旁腺可以被准确的切取 ,培养 5d的甲状旁腺细胞 ,细胞器最完整 ,分泌功能最强 ,可以作为移植的最佳供体

Abstract:Objective To study parathyroid gland’s excision technique and function in rat to establish foundation for parathyroid cell transplantation. Methods Parathyroid glands were excised by surgical operation and were identified by pathology. At the same time, parathyroid glands were digested with collagenase and trypsin, and then cells were cultured. The cells cultured for 5 days were observed by transmission electron microscopy, and the parathyroid hormone that was obtained from the cell fluids cultured for 1,3,5 and 7 days was measured by radioimmunoassay. Results Parathyroid glands from 18 rats were resected successfully, the resection rate was 90%(18/20). Abundant rough endoplasmic reticulaum,Golgi bodies,stab mitochondria,secretory granules and normal level of parathyroid hormone were found in the parathyroid cells cultured for 5 days. Conclusion Parathyroid glands in rats can be excised exactly,and the parathyrodi cells cultured for 5 days have maximum secreting function and optimum morphology and are optimal donors for transplantation.
