摘    要:目的 了解16型人乳头瘤病毒(HPV16)E6/E7、HSP70与角质形成细胞永生化及转化之间的关系。方法 Lipofectamine介导法进行转染,以G418筛选阳性克隆,免疫印迹分析及免疫荧光双标记染色结合激光扫描共聚焦显微镜对HSP70和HPV16E6/E7在转染然质形成细胞中的表达进行了观察。结果 免疫印迹分析显示正常负擀形成细胞和转染角质形成细胞均在相对分子质量70000处出现一条染色带

Expression of 70-kD Heat Shock Protein in Keratinocytes Induced by Human Papillomavirus 16 E6/E7

Abstract:Objective To investigate the relation between human papillomavirus 16 E6/E7 (HPV16 E6/E7),70 000 heat shock protein (HSP70) and immortalization and transformation of keratinocytes. Methods Keratinocytes were transfected with lipofectamine and positive clones were selected by G418. The expressions of HSP70 and HPV16 E6/E7 by transfected keratinocytes were analysed by means of Western blot,double labelling immunofluorescent staining and laser scanning confocal microscopy techniques. Results Results of Western blot analysis showed a specific band exhibiting a molecular weight of 70 000 in both normal keratinocytes and HPV16 E6/E7 transfected keratinocytes, but the staining was more intensive in transfected keratinocytes than in normal ones. The co location of HPV16 E6/E7 and HSP70 in transfected keratinocytes were observed by laser scanning confocal microscopy. Conclusion The high expression of HSP70 in transfected keratinocytes may be induced by HPV16 E6/E7 which suggests that HSP70 might play an important role in immortalization and even transformation of keratinocytes.
