摘    要:目的 研究多发性骨髓瘤(MM)中B淋巴细胞刺激因子受体(BAFF-R)基因、蛋白表达水平;并进一步研究BAFFR对MM细胞生长存活的影响.方法 应用RT-PCR,ELISA,Western Blot等技术检测BAFF-R在MM细胞中的表达;WST-1,TUNEL凋亡实验,Western Blot检测BAFF-R对MM细胞增殖、凋亡的影响.结果 BAFF-R mRNA和蛋白表达于MM细胞中;与对照组相比,BAFF-R阻断性抗体(1,5,15 μg/ml)可抑制KM3细胞增殖,t值分别为79.78,72.21,76.88;P值均为0.000,即BAFF-R促进KM3细胞增殖;同时,BAFF-R阻断性抗体组(15 μg/ml)细胞凋亡明显高于对照IgG组;BAFF-R阻断性抗体组抗凋亡蛋白Bcl-2表达降低18%,促凋亡蛋白Bax表达增加43%,说明BAFF-R能够抑制MM细胞凋亡.结论 BAFF-R对MM细胞生长存活有一定影响,BAFF-R对多发性骨髓瘤的发生发展发挥一定作用.

Study on the Expression of BAFF-R in Multiple Myeloma and Its Effect on Cell Survival

Abstract:Objective To investigate the expression of BAFF-R mRNA and protein in multiple myeloma and the effect of BAFF-R on cell survival.Methods BAFF-R mRNA and protein levels were detected by real-time PCR and Western Blot analysis,and the proliferation and apoptosis ability of MM cells were examined by WST-1,TUNEL assay and Western Blot.Results BAFF-R mRNA and protein were expressed in MM cells.The proliferation of MM cells treated with BAFF-R block antibody (1,5,15 μg/ml) were significantly more decreased than those in healthy controls,t values were 79.78,72.21,76.88,respectively (P<0.000 1).In other words,BAFF-R could induce the proliferation of MM cells.Meanwhile,there were significantly more apoptotic cells after treatment with BAFF-R block antibody (15 μg/ml) as compared with the control IgG.The results also showed that the expression of Bcl-2 protein was down-regulated by approximately 18 % and the expression of Bax protein was up-regulated by approximately 43% after inhibition of BAFF-R expression,suggesting that BAFF-R inhibited the apoptosis of MM cells.Conclusion BAFF-R may probably affect the growth of MM cells.The BAFFR expression may play a role in the carcinogensis and progress of multiple myeloma.
