骨质疏松模型大鼠骨量及血脂的变化

目的观察大鼠去卵巢30,60,90d后骨量及血脂的变化,验证雌激素变化与骨代谢及血脂代谢的时间、变化速度具有相关性的假设成立。方法42只大鼠单纯随机分为两组:假手术组和去卵巢组。分别在实验30,60,90d后处死,取血清及胫骨。观测骨密度(BMD)的变化并检测高密度脂蛋白胆固醇(HDL-C),低密度脂蛋白胆固醇(LDL-C)、总胆固醇(TC)、三酰甘油(TG)。结果大鼠去卵巢后体重增加的速度较快,60d后显著增加,平均为39.5g,比同期假手术组增加19.1g,90d后,同组比较,增加21.1g。去卵巢后,同期去卵巢组BMD均比假手术组减少(P<0.01,t=3.89,9.92,9.52),具有显著性差异,90d同30d相比,BMD从0.287g/cm2下降到0.245g/cm2,仍具有显著性(P<0.01,t=10.38)。30,60d后去卵巢组比同期假手术组LDL-C水平升高(P<0.05,t=2.57,2.46),90d组比较两组无显著性变化。HDL-C在整个实验过程中变化不大。30d组TC,TG水平均显著增加(P<0.01,t=3.48,3.10);60d组同期去卵巢组TC比假手术组增加7.3%(P<0.05,t=2.90),TG增加45.6%(P<0.01,t=3.05);90d组同期去卵巢组TC比假手术组增加6.1%,无统计学意义,TG增加45.6%,各假手术组的TC,TG,LDL-C水平随着时间的增加虽然有所增加,但不具有统计学意义。结论30d去卵巢大鼠已出现典型的骨质疏松表现,伴有血脂紊乱。60,

Changes of bone mass and serum lipid in rats with osteoporosis

Aim To study the changes of bone mass and b lood fat in rats with oste oporosis in 30days,60days,and 90days after operation for provability of the correlation betw een estrogen and the metabolism of bone and blood lipial.M ethods Forty-two female SD rats were equall y divided into two groups at rando m:sham opera tion group and ovariectomy group.Seven rats in the two groups were sacrificed at 30,60and 90days after the beginning of experime nt and their bl ood serum and tibias were taken out.The bone mass density(BMD ),high density lipoprotein choleste rol(HDL-C),low density lipoprotein cho lesterol(LDL-C),total cholesterol(TC)and triacylglycerol(TG)were measure d.Results The average body weight was 39.5g in the ovariectomy g roup and i ncreased 19.1g compared to that in the sham operation group at 60days afte r operation.The average body weight increased 21.1at 90days after operati on compared to that at 60days after operation.The BDM in the o variectomy gro up were lower than that in the sham operation group 30,60and90days after the beginning of ex-periment respectively,with signif icant differences betwee n the two groups(t=3.89,9.92,9.52,P<0.05).The BMD of the ovariectomy group reduced from 0.287g /cm 2 to 0.245g /cm 2 after 90days,compared to that after 30days(t=10.38,P<0.01 ).The levels of LDL-C in the ovariec-tomy group were higher than those in t he sham operation group at 30and 60days after operation(t=2.57,2.4 6,P<0.05),and there was no sig-nificant difference in the level of LDL -C between the two groups at 90days.The levels of TC and TG increased sign i ficantly in both groups at 60days after operation separately compare d with th ose at 30days after operatio n(t=3.48,3.10,P<0.01).The leve ls of TC and TG in the ovariec tomy group increased 7.3%and 45.6%more t han those in the sham operation group respectively 60days after operation.T he levels of TC and TG in the ovariectomy group increased 6.1%an d 45.6 %more than those in the sham operation group 90days after operat ion respecti vely.The levels of TC,T G and LDL-C in the sham operation group increased wit h time insignificantl y.Conclusion The ovariectomy rat shows the typical sympto m of osteoporosis is associated with blood lipid disorders 30days after operat ion an d inclines to a relative stationary p eriod until 60and 90days after operation.
