摘    要:目的:应用高速逆流色谱技术从连翘药材提取物中分离得到连翘苷。方法:采用两相溶剂系统,四氯化碳-氯仿-甲醇-水的四元体系,上相为固定相,下相为流动相;仪器转速为900rpm,流速为1.2mL.min-1。结果:经过ESI-MS和1H-NMR的鉴定,确认为连翘苷单体化合物,外标法测定纯度为98.8%。结论:本方法可以用于分离制备连翘苷单体。

Abstract:Phillyrin,an active component in the Fructus Forsythiae,was successfully separated from Chinese herbal medicine Fructus Forsythiae by High-Speed Counter-Current Chromatography(HSCCC).The solvent system used for separation was consisted of carbon tetrachloride-chloroform-methanol-water.The upper phase was used as stationary phase and the lower phase was used as mobile phase.The apparatus was rotated at 900 rpm and the mobile phase of solvent was pumped through the column at a flow rate of 1.2 mL/min.The identification by ESI-MS and 1H-NMR proved that the component was phillyrin,and the purity was 98.8%.It is concluded that HSCCC can be used in the separation of phillyrin.
