摘    要:目的:通过分析女贞子原植物及其混用品和伪品的ITS2序列,探究鉴定女贞及其混伪品的新方法。方法:采用CodonCode Aligner进行序列拼接,MEGA4.1计算女贞及其混伪品的种内、种间的K2P距离,并基于K2P模型构建NJ树,最后应用Schultz等建立的ITS2数据库和网站预测二级结构。结果:女贞的最小种间K2P距离(3.2%)大于种内最大K2P距离(0.90%),NJ树中女贞的不同来源样品聚为一支。女贞与其混伪品的二级结构的分子形态均有差异。结论I:TS2序列可以作为鉴定女贞子原植物及其混伪品的条形码序列,并且该序列在中药材原植物的鉴定中具有很大的潜力。

Abstract:Objective: To explore a new method to identify the original plant of Ligustri Lucidi Fructus from its adulterants by the ITS2 regions. Methods: Sequences were assembled using CodonCode Aligner. K2P distances were calculated and NJ Tree was performed by MEGA 4.1. Secondary structures were predicted by the ITS2 database and websites. Results: The minimum interspecific K2P distance (3.2%) was obviously higher than the maximum intraspecific one (0.90%). The different samples of Ligustrum lucidum were clustered into one clade. The molecular morphology of secondary structures of ITS2 was compared between Ligustrum lucidum and its adulterants. Conclusions:The ITS2 region could be used to discriminate the original plant of Ligustri Lucidi Fructus and its adulterants.
