梗阻性肾病大鼠肾脏肾上腺髓质素的表达及缬沙坦、红花对其的影响

摘    要:目的观察肾上腺髓质素在梗阻性肾病中的表达及缬沙坦、红花对其的影响。方法结扎SD大鼠单侧输尿管制备肾间质纤维化实验动物模型,同时给以缬沙坦及红花灌胃治疗,14d后摘取肾脏,免疫组化及原位杂交方法测定肾上腺髓质素的表达。结果梗阻性肾病实验动物肾脏肾上腺髓质素表达强度及面积均明显减弱,缬沙坦及红花可促进其表达。结论缬沙坦及红花拮抗肾间质纤维化可能与调控肾上腺髓质素的表达有关。

Abstract:Aim To investigate the expression of Adrenomedullin in obstructive nephropathy induced by unilateral ureteral obstruction (UUO) and treated with Valsartan and safflower. Methods SD rats were used to get the UUO and treated by Valsartan(10 mg·kg-1·d-1) and safflower(0.4 g·kg-1·d-1), the ligated kidneys were got at the 14th day after operation. The expression of adrnomedullin was determined with immunohistochemistry and in situ hybridization. Results The positive area and density of adrenomedullin was down-regulated in obstructive kidneys induced by UUO markedly and up-regulated by Valsartan and safflower. Conclusions Valsartan and safflower might inhibit the renal interstitial fibrosis through the adrnomedullin.
