细胞内游离钙在血管紧张素转换酶抑制剂逆转高血压病心脏重构中的作用

摘    要:目的 探讨细胞内游离钙在血管紧张素转换酶抑制剂 (ACEI)逆转高血压病心脏重构中的作用。方法 采用正常血压大鼠 (WKY)、自发性高血压大鼠 (SHR)配对 ,服药前后对比及离体淋巴细胞药物干预研究。结果 ①SHR与WKY比较有以下心脏重构现象 :心脏重量指数增加 ,心肌细胞肌节长度延长 ,肌束及核周水肿 ,胶原区扩大 ,心肌细胞内线粒体增多 ,体密度增加 ;线粒体肿胀 ,比表面减少 ,线粒体脊溶解和空泡样变性 ;②SHR外周淋巴细胞 Ca2 + ]i、血浆血管紧张素Ⅱ浓度 ATⅡ ]较WKY显著增加 ;③ACEI治疗后SHR心脏重构现象得到逆转 ,血浆 ATⅡ ]、淋巴细胞 Ca2 + ]i降低 ;④离体情况下 ,ATⅡ能使SHR、WKY淋巴细胞 Ca2 + ]i增加 ,ACEI能使淋巴细胞 Ca2 + ]i降低。结论 ACEI能有效逆转高血压心脏重构现象 ,其作用机制可能同有效纠正细胞游离钙超负荷及调节异常的细胞内游离钙代谢有关

Abstract:Objective To determine the relationship between the change of calcium metabolism and the effect of cardiac remodeling in SHR by ACEI therapy.Methods The changes in SHR and WKY before and after ACEI treatment,and the directly effect of calcium metabolism by ATⅡ and ACEI stimulation were observed.Results ①There was remodeling in heart of SHR:Cardiac indexes was increased;Mitochondrins got swollen and degenerat in cardiac cell;②The intralymphocytic free caluium concentrationCa 2+ ]i and were significantly higher in SHR;③After ACEI treatment,the blood pressure,Ca 2+ ]i and were decreased,the change of mitochondrins and the cardiac remodeling were reversed.④ATⅡ can directly increased intralymphocyticCa 2+ ]i,but ACEI can directly decreasedCa 2+ ]i in SHR and WKY.Conclusion The intracellular calcium changes may be involved to the hypotensive effect and the mechanism of reversing cardiac remodeling by ACEI tratment.
