利鲁唑对脂多糖诱导眼内炎中谷氨酸代谢的影响

摘    要:目的 观察利鲁唑对脂多糖诱导大鼠眼内炎中谷氨酸(glutamate,Glu)代谢的影响。方法 将SD大鼠随机分为3组,每组35只,1组为生理盐水对照组,2组为眼内炎组,3组为眼内炎利鲁唑干预组。2组、3组通过玻璃体内注射大肠杆菌脂多糖(lipopo-lysaccharide,LPS)建立大鼠眼内炎模型,3组腹腔注射利鲁唑进行干预。紫外分光光度计法检测玻璃体中Glu含量的变化,免疫组织化学方法测定谷氨酰胺合成酶(glutaminesynthetase,GS)在视网膜组织的表达变化及分布。结果 LPS诱导眼内炎后玻璃体内Glu浓度均显著增高。造模后6h玻璃体内Glu浓度即较对照组明显增加,1组玻璃体Glu浓度为(15.32±0.11)μmol·L-1,2组为(49.51±4.15)μmol·L-1,3组为(49.81±2.12)μmol·L-1,2组和3组差异无统计学意义,二者与1组相比差异均有统计学意义(均为P<0.05)。玻璃体内Glu浓度于48h达高峰后逐渐下降。2组在5d时Glu浓度较之前出现小幅回升(297.59±1.03)μmol·L-1,然后逐渐下降。6h后的各时间点,3组玻璃体Glu浓度均明显低于2组,差异均有统计学意义(均为P<0.05),但较1组仍明显增高(均为P<0.05)。GS在2组表达升高,主要表达于内颗粒层、神经节细胞层Müller细胞胞浆中。2组、3组在6h、12h、24h、48h、72h的GS表达差异均有统计学意义(均为P<0.05),3组GS的表达明显低于2组,但仍高于1组。结论 利鲁唑可以降低LPS诱导眼内炎中玻璃体Glu含量,抑制Müller细胞活化,提示其在细菌性眼内炎中具有保护作用。

Abstract:Objective To explore the effects of riluzole on glutamate( Glu) metabolism in endophthalmitis induced by lipopolysaccharide ( LPS) . Methods SpragueDawley rats were randomly divided int0 3 groups : BSS group( Group l) , LPS inoculated group ( Group 2) , LPS inoculated with riluzole treatment group ( Group 3 ) , 35 cases in each group. The right eyes of rats in group 2 and group 3 were injected with LPS. and the BSS rats received intravitreal injections of balanced salt solution , respectively. Intraperitoneal injections of riluzole were given to the rats in group 3 before and after LPS inoculation , once daily during the next three days. The levels of Glu in the vitreous humor and retinal glutamine synthetase ( GS) expression were investigated. Results The concentration of Glu increased after endophthalmitis induced by LPS. The levels of Glu in vitreous of group l,2,3 at 6 hours after modeling were ( 15. 32 + 0. 11) ymol . L-l , (49. 51 + 4. 15 ) Vmol . L -l and ( 49. 81 + 2. 12 ) p mol . L -1 , respectively, there was no statistical difference between group 2 and group 3 ,but there were statistical differences between above two groups and group I ( both P < 0. 05 ) . The concentration of Glu in the vitreous humor peaked at 48 hours , then gradually decreased, group 2 slightly increased at 5 days , then gradually decreased. At each time points after 6 hours , the concentration of Glu in the vitreous humor of group 3 were all lower than those of group 2 ( all P < 0. 05 ) .but were higher than those of group I ( all P < 0. 05 ) . The expression of GS in group 2 increased. which mainly expressed in ganglion layer and inner nuclear layer. There were statistical differences in the expression of GS at 6 hours . 12 hours ,24 hours. 48 hours ,72 hours between group 2 and group 3 ( all P < 0. 05 ) , the group 3 were lower than group 2, but still higher than group l. Conclusion Riluzole can decrease the content of Glu in the vitreous humor of endophthalmitis induced by LPS, inhibit the muller cells activation,which suggests that riluzole may play the protective role in the endopthamitis.
