乳腺癌组织Livin和PTEN的表达及与肿瘤细胞凋亡的相关性

摘    要: 目的研究乳腺癌组织中凋亡抑制蛋白Livin和抑癌基因PTEN的表达与肿瘤细胞凋亡的关系,探讨两者在乳腺癌细胞凋亡、侵袭转移中的作用机制。 方法应用免疫组化Elivision法检测90例乳腺癌和30例乳腺纤维腺瘤组织中Livin和PTEN的表达,另以TUNEL法检测细胞凋亡情况,分析Livin和PTEN表达与乳腺癌细胞凋亡的关系。 结果在90例乳腺癌中Livin和PTEN的表达阳性率分别为54.4%, 48.9%,凋亡指数为4.79%±1.62%,与良性对照组比较均有显著差异(P < 0.01);Livin阳性表达与乳腺癌肿瘤大小、临床分期呈正相关(P < 0.05),与凋亡指数呈负相关(P < 0.05),与年龄、病理分级、淋巴结转移未见显著相关(P>0.05);PTEN表达与乳腺癌淋巴结转移、临床分期呈负相关(P < 0.05),与凋亡指数呈正相关(P < 0.05),与年龄、肿瘤大小、病理分级未见显著相关(P>0.05)。 结论Livin和PTEN对乳腺癌细胞凋亡分别起抑制和促进作用,Livin高表达和PTEN失表达都可通过抑制细胞凋亡促进乳腺癌的侵袭转移,调控两种基因的表达有望影响乳腺癌细胞凋亡以及乳腺癌的发生发展。

The expression of Livin,PTEN and their relationships with apoptosis in breast cancer

Abstract: Objective To investigate the expression of inhibitor of apoptosis protein Livin and tumor suppressor gene PTEN and its association with tumor apoptosis of breast cancer, and to study the mechanism of apoptosis, invasion and metastasis in breast cancer. Methods Expressions of Livin and PTEN were examined by Elivision immunohistochemistry in 90 cases of breast cancer tissues and 30 cases of breast fibroadenoma tissues. The apoptosis in breast tissues was examined by TUNEL. To analyze the relationships between expression of Livin and PTEN and apoptosis of breast cancer. Results In 90 cases of breast cancer, the expression positive incidences of Livin and PTEN were respectively 54.4%, 48.9%, the apoptosis index (AI) was 4.79%±1.62%, which all differentiate apparently those of the benign control group (P < 0.01). In breast cancer tissues, the expression of Livin was positively related to tumor size, clinical stage (P < 0.05), but negatively related to AI (P < 0.05), but unapparently related to patients age, histopathologic grade, lymphnode metastasis (P>0.05). The expression of PTEN was negatively related to lymphnode metastasis, clinical stage (P < 0.05), but positively related to AI (P < 0.05), but unapparently related to patients age, tumor size, histopathologic grade (P>0.05). Conclusion Livin and PTEN respectively play the roles of inhibiting and promoting in apoptosis of breast cancer. Both the high expression of Livin and the loss expression of PTEN can promote invasion and metastasis by restraining apoptosis in breast cancer. It is hopeful to effect apoptosis, carcinogenesis and development through regulating Livin and PTEN expression in breast cancer.
