高迁移率族蛋白B1在肿瘤发生发展中的作用

摘    要:高迁移率族蛋白B1(high mobility group box 1, HMGB1)是一种非组蛋白细胞核蛋白,属于损伤相关分子模式(DAMPs)中的一员,在多种肿瘤组织中高表达。可增强肿瘤细胞的增殖、侵袭和转移,促进肿瘤的发生发展;也可增强免疫反应,诱导肿瘤细胞凋亡,抑制肿瘤的发生发展;提示HMGB1在肿瘤中扮演着“双刃剑”的角色。这主要与HMGB1的定位和翻译后修饰有密切关系。本文就与肿瘤发生发展相关的HMGB1结构、翻译后修饰、定位、功能及目前HMGB1在各个肿瘤中概况作一综述。

Abstract:High-mobility group Box-1(HMGB1) is a non-histone nuclear protein and a member of damageassociated molecular pattern molecules(DAMPs). High expression of HMGB1 in tumor tissues could increase cell proliferation, invasion and migration, and then improve the tumorigenesis; on the contrary, studies also indicated that high level of HMGB1 could inhibit tumorigenesis in the way of inducing tumor cell apoptosis and enhancing the immunoreaction. These insights imply that HMGB1 plays a “double-edged sword” role in the tumorigenesis, which is highly related to the location and post-translational modification of HMGB1. In this article, we will review the basic characteristics, post-translational modification, location and function of HMGB1 and the research advances of HMGB1 in many tumors.
