复方北豆根提取物胶体溶液的制备及抗人乳头瘤病毒的实验研究

 目的探讨复方北豆根提取物胶体溶液抗人乳头瘤病毒(human papillomavirus,HPV)的分子机制,研究中药胶体溶液制备工艺流程,开发出适合女性HPV感染的新型药物释放系统。方法从北豆根中提取总碱,从蚕蛹中提取复合氨基酸,与铜绿等矿物质络合成胶体溶液,采用荧光定量PCR技术检测胶体溶液对离体尖锐湿疣中HPV-DNA复制载量。结果复方北豆根提取物胶体溶液(1 g·mL^-1)及0.1倍稀释药物使HPV6/11和HPV16/18型DNA复制载量降为(1.5±1.0)copies·mL^-1,与阴性对照组HPV-DNA复制载量比较,差异有统计学意义(P<0.01)。结论北豆根总碱与蚕蛹复合氨基酸、铜绿等矿物质络合成复方北豆根提取物胶体溶液可抑制HPV-DNA复制,具有明显的高活性抗HPV作用。

Preparation of the Colloid Solution of Compound Menispermum dauricum Extract and Its in Vitro Pharmacodynamics Effect on Anti-Human Papollomavirus

OBJECTIVE To develop the new kind of drug-releasing system containing colloid solution of compound Menispermum dauricum extract(CMDE) for the treatment and prevention of HPV,and to explore the molecular mechanism of anti-HPV.METHODS The colloid solution containing total alkaloids of Menspermum dauricum,bombyxmoril′s compound amino acids and minerals including Cu₂(OH)₂CO₃ and borax was prepared.The fluorogenic quantitative PCR was used to detect the antiviral effect by determining the HPV-DNA amplification.RESULTS The colloid solution of CMDE(1 g·mL^-1) and 10^-1 diluted drug(0.1 g·mL^-1)greatly inhibited the quantity of HPV-DNA with the amplification of(1.5±1.0) copies·mL^-1.Compared with the negative control groups,there were signicant difference between the two groups(P<0.01).CONCLUSION The colloid solution of CMDE had a great potential anti-HPV by decreasing the duplication of HPV.
