缺氧条件下缺氧诱导因子-1α对人舌鳞癌细胞血管内皮生长因子表达的影响

摘    要:目的探讨缺氧条件下缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-1α)对人舌鳞癌细胞株Tca8113细胞血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法采用RT—PCR技术半定量检测人Tca8113细胞常氧和缺氧培养0.5、1、3、6、12和24h HIF-1α、VEGF基因的表达。结果缺氧条件下HIF—1α基因的表达呈先升后降的趋势,在缺氧6、12h表达显著高于常氧组(P〈0.05);VEGF的表达在缺氧培养0.5、1、3、12及24h均高于常氧组(P〈0.05);VEGF基因的表达在缺氧初始阶段随HIF—1α基因表达的增强而升高,但Spearman相关分析显示两者在缺氧24h内相关性不明显(rs=0.5750,P〉0.05)。结论缺氧条件下Tca8113细胞中VEGF的高表达受HIF—1α的调控和影响,是多种因素作用的结果。

Abstract:OBJECTIVE: To investigate the effect of hypoxia inducible factor-1alpha (HIF-1alpha) on vascular endothelial growth factor (VEGF) expression in Tca8113 cells under hypoxia. METHODS: The expression of the mRNA of HIF-1alpha and VEGF in Tca8113 cells was examined by RT-PCR technique at different culture times (1/2 h, 1 h, 3 h, 6 h, 12 h, 24 h) under normoxic and hypoxic conditions. RESULTS: The expression of HIF-1alpha under hypoxia showed the trend of increasing first and then decreasing, and was higher than that of the control (normoxic group) at 6h and 12 h (P < 0.05). The expression of VEGF under hypoxia was higher than that of the control group at 1/2 h, 1 h, 3 h, 12 h, 24 h (P < 0.05). The expression of hypoxia-induced VEGF mRNA increased with the increased expression of HIF-1alpha mRNA in the cell lines tested at the initial stage of hypoxia. But no statistical significant association was observed between HIF-1alpha and VEGF expression within 24 h under hypoxia (rs = 0.5750, P > .005). CONCLUSIONS: The increased expression of VEGF in Tca8113 cells might be mediated by multiple factors, including HIF-1alpha.
