甘草地上部分水提液总黄酮纯化工艺研究

摘    要:目的研究大孔树脂纯化甘草地上部分水提液中总黄酮的工艺。方法通过对D-101、AB-8、DM-130、HPD-BJQH、HPD-600、HPD-100和HPD-450等7种树脂对总黄酮的静态吸附与解吸附考察,优选适宜类型的大孔树脂,并进一步对富集纯化参数进行考察及优化。结果选用AB-8型大孔树脂,优选的工艺条件为:上样液质量浓度2.128 mg/m L,样品液p H5.42,树脂的上样量为7 m L/g,上样速度为1.5 BV,依次用2 BV水洗脱,4 BV 60%乙醇洗脱,经AB-8树脂处理后的甘草地上部分水提液的总黄酮纯度由9.08%提升到34.02%,收率为74.40%。结论 AB-8型大孔树脂能有效地纯化富集甘草地上部分水提液总黄酮,且此工艺稳定。

Abstract:Objective To investigate the purification technology of total flavonoids of water extract from the aerial parts of licorice.Methods Seven macroporous resins (D-101、AB-8、DM-130、HPD-600、HPD-100 and HPD-450) were chosen with static and dynamic adsorption and desorption experiments to optimize the purification parameters.Result AB-8 macroporous resin was chosen and its optimized technology conditions were as followings:the concentration of sample solution 2.128 mg/mL(pH=5.42),the loaded amount of 7mL/g,and loading flow rate of 1.5 BV/h.The sample was eluted by distilled water with 2 BV and 60% ethanol of 4BV,respectively.The purity of total flavonoids was increased from 9.08% to 34.02% after the purification,and the yield was 74.40%.Conclusion AB-8 is an ideal resin with the best enrichment for separating and purifying the total flavonoids of water extract from the aerial parts of licorice.
