肾阳虚血瘀证子宫内膜异位症大鼠造模方法的建立与评价

摘    要:目的:研制肾阳虚血瘀证子宫内膜异位症(EMT)病证结合大鼠模型,以供EMT中医药实验研究应用。方法:模拟妇科"冒雨涉水,感受寒邪"的病因,依据"寒邪伤阳"的中医理论,与传统的"自体移植"EMT动物模型相结合,建立病证结合的肾阳虚血瘀证EMT动物模型。依据冷冻时间长短及前后顺序行3种不同的造模模式。冷冻1个月,行EMT造模,再冷冻1个月设为"冷冻+造模+冷冻"组;冷冻1个月,行EMT造模后不冷冻为"冷冻+模型"组;先不冷冻,行EMT造模后,再冷冻1个月,设为"造模+冷冻"组,另设假手术组。用理化检验指标评价该模型的临床符合度。结果:与冷冻+模型组、造模+冷冻组、假手术组比较,冷冻+造模+冷冻组EMT的大鼠模型能反映肾阳虚血瘀证的中医基本证候特征,与EMT的疾病特点相符合。结论:冷冻+造模+冷冻方法是理想的肾阳虚血瘀证EMT实验模型。

Abstract:Objective: to develop the models with endometriosis (EMT) of kidney yang deficiency and blood stasis syndrome for the study and application of EMT in TCM. Methods: simulating the cause of "wading in the rain and getting the cold evil" in gynecology, EMT models with kidney yang deficiency and blood stasis syndrome were established on the basis of TCM theory "pathogenic cold impairing yang", in combination with traditional "autotransplantation" EMT animal model. Three different models were set up according to the time and the sequence of freezing time. Frozen for 1 month, EMT models were made before being frozen for another month for "freezing+molding+freezing" group;frozen for 1 month, EMT models were made for "freezing+molding" group;EMT models were directly made before being frozen for 1 month for "molding+freezing" group;and the sham-operated group was also established. Physical and chemical indexes were used to evaluate the clinical compliance of the models. Results: compared to the other three groups, the models of "freezing+molding+freezing" group could reflect the TCM syndrome characteristics of kidney yang deficiency and blood stasis syndrome, which was consistent with the disease characteristics of EMT. Conclusion: freezing+molding+freezing method is the ideal experimental models for EMT with kidney yang deficiency and blood stasis syndrome.
