摘    要:目的:探讨黄芪总提物(TAE)对荷瘤小鼠化疗引起的白细胞减少和免疫抑制以及抗肿瘤作用的影响。方法:用环磷酰胺(CTX)或5-氟尿嘧啶(5-Fu)致正常与荷瘤小鼠的白细胞减少或免疫抑制模型,观察TAE对外周血白细胞、胸腺指数及血清IL-2的影响。结果:TAE(15、50和150mg/kg)与CTX(80mg/kg,腹腔注射(ip),第6、7天)合用,可明显提升CTX降低的小鼠外周血白细胞。TAE与CTX(50mg/kg,ip,第1、4、7、10天)合用治疗荷S180小鼠,在保持CTX疗效的同时,对其降低外周血白细胞数,亦有明显的提升作用。TAE与5Fu合用,可使5Fu降低的HepA小鼠胸腺指数和血清IL2含量上升。结论:TAE对化疗引起的白细胞减少和免疫抑制有明显的保护作用,且对其抗肿瘤活性有一定的增效作用。

Abstract:Objective: To investigate whether total astragalus extralct(TAE),could protect against chemotherapy-induced leukopenia and immunosuppression,and whether it affects Cyclophosphamide(CTX) or 5-Fluorouracil(5-Fu) antitumor activity. Method: 5-Fu-induced immunosuppressed model in HepA tumor bearing mice and CTX-induced leukopenia in normol and S180 bearing mice were established. Result: CTX markedly decreased WBC count of normal and S180 bearing mice ,all the above leukopenia were reduced significantly by TAE. Immunosuppression of 5-Fu in mice bearing with HepA as measured by decreases of the thymus index and serum interlukin-2(IL-2 )level was significantly alleviated by TAE. Moreover,the antitumor activity of CTX or 5-Fu ,as assayed by tumor weight of mice bearing with S180 or HepA was not reduced or was increased by TAE treatment.Conclusion: TAE may protect immune system and bone marrow against chemotherapy-induced toxicity with increasing or without interfering with their antitumor activity in mice.
