都匀亚洲牛带绦虫病小鼠肝纤维化肝脏CTCF、TCF-β1的表达意义

摘    要:目的研究结缔组织生长因子(connective tissue growth factor,CTGF)与转化生长因子-β1(transforming growthfactor-betal,TGF-β1)在都匀亚洲牛带绦虫小鼠肝纤维化肝脏中的表达状况及意义。方法用昆明小鼠感染都匀亚洲牛带绦虫建立小鼠肝纤维化模型;感染小鼠于30、40、50、60 d剖杀,HE染色观察肝脏病理变化,免疫组化法检测肝内CTGF、TGF-β1蛋白的表达水平。结果感染小鼠30 d时形成都匀亚洲牛带绦虫肝纤维化,40d时肝内CTGF和TGF-β1蛋白表达达高峰,随后出现下降,感染小鼠CTGF和TGF-β1蛋白表达水平具有直线相关性。结论CTGF与TGF-β1的表达与都匀亚洲牛带绦虫小鼠肝纤维化形成有密切关系,TGF-β1的致纤维化作用可能部分通过CTGF的生物学作用介导,阻断CTGF的传导通路可能是绦虫病肝纤维化治疗的有效靶点。

The expression and significance of CTCF and TCF-β1 in murine liver fibrosis induced by Taenia Saginata Asiatic from Duyun area

Abstract:Objective:To study the expression and significance of connective tissue growth factor (CTGF) and transforming growth factor-batal (TGFβ1) in murine liver fibrosis induced by Taenia saginata Asiatic from Duyun area. Methods: Establish the murine liver fibrosis model induced by Taenia saginata Asiatic from Duyun area, and subsequently killed at the 30^th,40^th,50^th. and 60^th days respectively. The hepatic expression of CTGF and TGF-β1 was detected by immunohistochemical method. Histological changes of murine liver were observed by HE staining. Results: The routine liver fibrosis model induced by Taenia saginata Asiatic from Duyun area were successfully established at the 30^th day. The hepatic expression of CTGF and TGF-β1, in the murine liver fibrosis models peaked at the 40^th day and then declined. There existed positive correlation between the hepatic expression of CTGF and TGF-β1. Conclusion: The expression of CTGF and TGF-β1 is closely correlated to the development of the murine liver fibrosis induced by Taenia saginata Asiatic from Duyun area, and TGF-β1 may play its fibering roles by roles action on CTGF. To block the CTGF signal transduction pathway may be a potential treatment of Taenia Saginata Asiatic liver fibrosis.
