THE CHANGES OF SOME BIOLOGICAL PROPERTIES OF HERPES SIMPLEX VIRUS TYPE 2 IRRADIATED QUANTITATIVELY BY ULTRAVIOLET LIGHT
Abstract:The strain wu of herpes simplex virus type 2 (HSV-2w) was irradiated with a coil tube lamp of ultraviolet light at an average dose of 39.9ergs/sec/mm~2 for different time and then inoculated into the monolayer cultures of rabbit kidney cells.The changes of some biological properties of HSV-2w were studied. When HSV-2w was irradiated for 2 minutes,its TCID_(50)/0.1ml and PFU/ ml decreased respectively from 10~(-3·3) to 10~(-2·3) and from 1.6×10~5 to 3.0×10~3. But the multinuclear cells were still present.With the electron microscope, a few of swelling and degenerating mitochontria was found in the cells infected by the virus.The virus particles were recognized in the intercellu- lar spaces,cytoplasm and lysosomes.No formation of the nucleocapsids could be found in the nucleus while a lot of them was present in the nucleus of cells infected with the unirradiated HSV-2w. When irradiating for 4 minutes,the number of TCIL_(50)/0.1ml and PFU/ ml of the virus was zero.Additionally,the monolayer cultures of rat embryo lung cells infected with the virus irradiated for 4 minutes seemed to become more easily passaged in vitro than those infected with the unirradiated virus. With the electron microscope,only a few of virus particles could be recog- nized inside and outside the infected rabbit kidney cells,but the nucleocap- sids were never observed in the nucleus of the cells.The number of the or- ganells in the cells was diminished and the amount of the microfilaments was increased. When the virus was irradiated for 6 minutes,no CPE and virus particles could be found in the cytoplasm.But a few granules of the virus-specific antigens was still demonstrated by the immunoperioxidase technique. The data presented in this experiment suggested that the irradiation of HSV-2w(10~5PFU/ml) with ultraviolet light at an average dose of 39.9ergs/ sec/mm~2 for 4~6 minutes could partially inactivate the viruses and lead to a change of the productive infection into the non-productive infection in the monolayer cultures of the rabbit kidney cells.
