摘    要:目的:制作大鼠颈部肺动脉高压动物模型。方法:将雄性Wistar大鼠随机分为实验组(n=15)熏对照组(n=10)。实验组大鼠右颈总动脉和颈外静脉用套管连接法连接。术后满16周测大鼠右心室及肺动脉压力,对肺组织行形态学观察。结果:实验组大鼠肺动脉和右心室收缩压与对照组比较差异有统计学意义(P<0.001);实验组大鼠右心室/体重及右心室/(左心室 室间隔)与对照组比较差异有统计学意义(P<0.001);组织切片见实验组大鼠肺动脉内膜层及肌层增厚,管腔狭窄。结论:套管连接法可成功制作大鼠左向右分流肺动脉高压模型。

Abstract:Objectives: A pulmonary hypertension model in male Wistar rats undergone cervical left to right shunt were developed by using a cuff technique connecting the left external jugular vein with left common carotid artery. Methods: 15 rats undergone the above surgery were selected as experiment group (n=15), and 10 male rats were selected as control group (n=10).16 weeks after the surgery, the right ventricle systolic pressures (sRVP) and pulmonary artery systolic pressures(sPAP) of rats were measured, and the morphology changes of the rats lung tissues were observed. Results: the pulmonary artery and right ventricle pressures are significantly increased in experiment group in contrast with control group (P<0.001); the ratios of right ventricle to body weight (R/BW) and right ventricle to left ventricle plus septal (R/(L S)) are also demonstrated statistically different (P<0.001). However, the ratios of double lung to body weight and the body weight do not express any difference in two group rats, (P>0.05). Lung tissue staining shows that the pulmonary artery walls are thickened, especially in the middle layer in experiment group rats. Conclusions: Development of this new rat cervical left to right shunt pulmonary hypertension model by using cuff technique provide an ideal and simple animal model for further investigating the molecule and biology mechanisms and drug interference effect of left to right shunt pulmonary hypertension.
