骨髓基质干细胞不同移植方式治疗脊髓损伤的实验研究

摘    要:目的探讨MSCs不同的移植方法对脊髓损伤修复的影响.方法90只成年大鼠,随机分为对照组、静脉移植组和腹腔移植组(每组再分为1、3、7、14、21d组),.在脊髓损伤后2d,经腹腔及静脉移植带绿色荧光的MSCs,,大鼠于移植后1、3、7、14、21d分批处死.连续冰冻切片观察细胞是否能够通过觑脑屏障并迁移到损伤部位,同时用体视学方法观察MSCs在脊髓损伤与非损伤部位的密度,用BBB评分法和爬网格法观察大鼠后肢运动功能的恢复.结果静脉移植后第1天,在损伤部位仅有少量的MSCs,而腹腔移植组未见MSCs。第3天有少量的MSCs.腹腔移植组与静脉移植相比7,14d有显著差异(P〈0.05).21d后两种移植方法在损伤部位的细胞无差异(P〉0.05).BBB评分,移植后7d无差异,14d后有明显差异(P〈0.05).脊髓损伤处的细胞密度明显多于非损伤部位(P〈0.05).结论MSCs经静脉和腹腔移植均能透过血脑屏障迁移到脊髓损伤部位并对脊髓损伤有一定的修复作用,静脉移植优于腹腔移植.

Abstract:Objective To observe the influence of the different methods of transplanting MSCs to cure spinal cord injury.Methods Ninety adult srague-daweley rats were used and randomly divided into three groups: control groups,intravenous transplantation and abdominal cavity transplantation groups(Every group was re-divided: 1-day group,3-day group,7-day group,14-day group,21-day group).2 days after injury,MSCs with green fluorescence were transplanted by intravenous and abdominal cavity transplantation.Then the rats were killed after 1,3,7,14,21 days to observe whether the MSCs can move to injury location through blood-brain barrier.And the density of MSC was detected by serology in injury and non-injury area.The locomotor function of the rats was measured by BBB score and climbing onto in lined grids.Results One day after intravenous transplantation,there were a few MSCs in the injury spinal cord location,but through abdominal cavity transplantation,the MSCs were not detected,and 3 days later,there were a few.7 and 14 days later,comparison between abdominal cavity transplantation and intravenous transplantation showed significant difference(P<0.05).21 days later,there was no difference between the two groups(P>0.05).Assessed by BBB score,there was no difference within control groups in 7 days.14 days later,there was significant difference within the three groups.The MSC density was larger in the injuries than in non-injury area.Conclusion MSCs are able to survive and move to injury spinal cord location through abdominal cavity and intravenous transplantation.MSCs may provide an autoplastic therapy for spinal cord injury,and intravenous transplantation was better.
