肿瘤患者红细胞免疫功能与SAPO-1/Fas、TNF-a、NO表达水平的实验研究

摘    要:目的:为了探讨肿瘤患者红细胞免疫功能与SAPO-1/Fas、TNF-a、NO表达水平的相关性及相互作用。方法:采用酵母菌-红细胞免疫粘附试验,对45例化疗前和17例化疗后病情缓解者进行了红细胞C3b受体花环试验(RBC-C3bRR)和红细胞免疫复合物花环试验(RBC-ICR)及ELISA法对107例肿瘤患者及22例健康人进行了SAPO-1/Fas的检测,其中部分进行了TNF-a、NO检测。结果:肿瘤患者血清SAPO-1/Fas表达水平明显高于健康人对照组(P<0.001),且与肿瘤种类无关;17例化疗后病情缓解者SAPO-1/Fas表达水平与化疗前显著降低(P<0.05)。45例和47例肿瘤患者血清同时检测SAPO-1/Fas、TNF-a表达水平及SAPO-1/Fas、NO含量并进行单因素直线相关分析,分别为r=0.76,P<0.05,r=0.72,P<0.05,呈显著正相关。RBC-C3bRR明显降低,45例肿瘤患者化疗前又明显低于17例病情缓解者(P<0.01),RBC-ICR明显高于健康人,45例化疗前肿瘤患者RBC-ICR明显高于17例化疗后病情缓解者(P<0.01)。结论:肿瘤患者RBC-ICR和SAPO-1/Fas、TNF-a、NO表达水平明显增高,呈显著正相关,RBC-C3bRR明显降低。为研究红细胞免疫与淋巴细胞免疫的相互关系及肿瘤治疗提供了新的理论基础和实验依据。

Abstract:Objective:To investigate the correlation and interactions between erythrocyte immune function and levels of SAPO-1/Fas,TNF-a and NO in tumor patients.Methods: Yeast-erythrocytoimmune adherence test was used to detect the levels of red blood cell C3b receptor rosette(RBC-C3bRR) and immune complex rosette(RBC-ICR) in 45 tumor patients before chemotherapy and 17 response cases after chemotherapy.ELISA was used to detect the levels of SAPO-1/Fas in 107 Tumor Patients and 22 controls,and the levels of TNF-a and NO in some cases.Results: Levels of SAPO-1/Fas in serum of tumor patients were obviously higher than those of controls(P<0.001),irrespective of whatever tumor.Levels of SAPO-1/Fas in 17 response cases after chemotherapy were obviously lower than those before chemotherapy(P<0.05).Levels of SAPO-1/Fas and TNF-a in 45 tumor patients and levels of SAPO-1/Fas and NO in 47 tumor patients were detected respectively.Univariate linear correlation analysis was used for comparison among the variables,which showed all had obvious association(r=0.76,P<0.05 and r=0.72,P<0.05 respectively).RBC-C3bRR levels were obviously reduced,especially lower in 45 tumor patents before chemotherapy than those in 17 response cases after chemotherapy(P<0.01).RBC-ICR levels in tumor patients were more increased than those in the controls,especially higher in 45 tumor patients before chemotherapy than those in 17 response cases after chemotherapy(P<0.01).Conclusion: In tumor patients levels of RBC-ICR,SAPO-1/Fas,TNF-a and NO were obviously increased and had obvious association,level of RBC-C3bRR was obviously reduced.This study provided a new theory basics and experimental clue for the interactions between erythrocyto immune and lymphocyte immune and tumor therapy.
