VEGF 3种受体在胶原性关节炎大鼠关节的表达与软骨破坏的相关性分析

检测血管内皮生长因子特异性受体在胶原性关节炎滑膜表达，并探讨其表达与关节软骨含量的相关性。采用RT—PCR方法检测VEGFR—1、VEGFR—2、VEGFR—3 mRNA表达，用GAPDH内参照相对定量VEGFR mRNA表达丰度。发现VEGFR—1在胶原性关节炎和对照组表达无显著差异；VEGFR—2、VEGFR—3在胶原性关节炎和对照组表达有显著差异。VEGFR—2、VEGFR—3 mRNA表达丰度与胶原性关节炎软骨含量呈负相关。提示VEGFR—2、VEGFR—3与类风湿关节炎软骨破坏有关，特异性阻断VEGFR—2、VEGFR—3可能改善类风湿关节炎预后。

Expression of VEGF-receptors in collagen-induced arthritis of rodent and their correlations with articular cartilage destruction

To identify the expression of specific receptors of vascular endothelial growth factors in synovial membrane of collagen induced arthritis of rodent, and correlate the expression with concentration of articular cartilage. RT PCR was used to detect VEGF receptors expression. Semi quantification of VEGFRs mRNA expression was done by comparing VEGFRs mRNA with internal control GAPDH. No statistical difference between VEGFR 1 mRNA expression in CIA and control group was found. Significant difference was observed between VEGFR 1 mRNA expression in CIA and control group.VEGFR 2 and VEGFR 3 mRNA expression were negatively correlated with articular cartilage concentration. It implies that VEGFR 2 and VEGFR 3 might be involved in articular cartilage destruction in RA/CIA. Targeting VEGFR 2 and/or VEGFR 3 may be a strategy to prevent articular cartilage destruction in RA/CIA.
