摘    要:目的 采用高效液相色谱法研究并建立牦牛皮胶氨基酸类成分的指纹图谱,为牦牛皮胶的质量控制提供有效的方法.方法 采用柱前衍生-RP高效液相色谱法,梯度洗脱进行分离,固定相为Agilent Analytical XDB-C18,(4.6 mm ×250mm,5μm);流动相为pH6.4的醋酸钠缓冲液(A) - 60%乙腈(B),检测波长为360 nm,体积流量为1.0 mL/min,分析时间为48 min.结果 10批样品具有较高的相似度.天冬氨酸、谷氨酸、丝氨酸、甘氨酸、苏氨酸、脯氨酸、丙氨酸、缬氨酸、甲硫氨酸、异亮氨酸、亮氨酸、苯丙氨酸和赖氨酸的平均质量分数分别为5.58%、10.49%、3.35%、23.25%、1.74%、14.19%、8.98%、2.48%、0.29%、1.6%、3.07%、2.12%和3.57%.结论 该方法稳定可靠,可为其氨基酸质量的全面控制提供参考.

Abstract:AIM To develop an HPLC method for controlling the quality of yak collagen according to the amino acids constituents.METHODS The amino acids constituents were analysed by Pre-column derivatization reversed phase high performance liquid chromatography.Chromatographic conditions were as follows: Agilent Analytical XDB-C18 column(4.6 mm×250 mm,5 μm),mobile phase was natrium aceticum buffer solution(A)(pH = 6.5)-60%acetonitrie(B),the detective wavelength was set at 360 nm,the flow rate was 1 mL /min and the analytical time was 48 minutes.RESULTS The result showed that the amino acids constituents had a high similarity.The contents of Asp,Glu,Ser,Gly,Thr,Arg,Ala,Val,Met,Ile,Leu,Phe and Lys were 5.58%,10.49%,3.35%,23.25%,1.74%,14.19%,8.98%,2.48%,0.29%,1.6%,3.07%,2.12% and 3.57%,respectively.CONCLUSION The method has good reproducibility and stability,and can be served as the means to control the quality of yak collagen.
