肝癌中PTEN基因及蛋白表达的意义

目的:研究肝癌中PTEN基因mRNA和蛋白的表达与肝癌病理之间的关系。方法:应用RT-PCR法检测28例肝癌组织和对应的癌旁组织中PTEN基因mRNA表达,并通过电泳凝胶图像系统分析其相对表达量。同时采用免疫组化方法检验51例肝癌石蜡切片中PTEN蛋白表达(其中包括与28例新鲜组织相对应的标本)。结果:RT-PCR检测PTENmRNA表达率肝癌中为35.71%,癌旁组织中为71.43%,PTEN基因在肝癌细胞中表达显著低于癌旁肝组织。免疫组化检测肝癌中PTEN蛋白的检出率为66.7%,PTEN蛋白表达与肝癌的病理分级和癌栓形成呈负相关。结论:PTEN基因缺失和肝癌的分化程度以及癌栓形成、转移密切相关。

Significance of PTEN Gene and Protein Expression in Hepatocellular Carcinoma

Objective:To investigate PTEN gene and protein expression in HCC and peritumor tissue,compare their difference and clinicopathological significance.Method:PTEN mRNA expression was detected by RT-PCR,28 cases of HCC and peritumor tissue are examined.Four-starimage analysis system was used to check relative expression quantity.PTEN protein expression is examined by immunohistochemistry,51 cases of HCC are detected,which consists of 28 cases of fresh tissues′counterpart.Results:The positive expression rate of PTEN gene mRNA is 35.71% in HCC,71.43% in peritumor tissue,mRNA expression rate is signifcant lower in HCC.The positive expression rate of PTEN protein is 47.06%,PTEN protein expression intensity is closely associated with pathological grading(P=0.0179) and vein metastasis(P=0.0388).Conclusion:PTEN gene expression is closely associated with occurrence and vein metastasis of HCC.
